Objective: To investigate and compare the content of SDF-1α protein in peripheral blood of gastric cancer and its clinical significance.Materials and Methods: The contents of SDF-1α in peripheral blood of 90 patients and25 healthy volunteers were detected by enzyme-linked immunosorbent assay. The correlation of SDF-1α content with the clinicopathologic parameters and prognosis after operation were analyzed at the same time. The independent risk factors of SDF-1α high content were analyzed by Logistic regression. The independent risk factors of gastric cancer patients’ prognosis were analyzed by COX’ s proportional hazard regression.Results: the content level of SDF-1α in patients with tumor(n=90) [(6950.8 ±1131.3)pg/ml] was significantly higher than that in control normal healthy volunteers(n=25)[(5023.7 ± 1103.8)pg/ml,(P=0.036) ]. The content of SDF-1α in gastric cancer patients(n=12)[(8251.6 ± 1042.5)pg/ml] with distant metastases was significantly higher than that without distant metastases(n=78)[(6785.3 ± 1025.0)pg/ml,(P<0.001) ]. The higher content of SDF-1α in peripheral blood was correlated with later TNM stage(P<0.001), broader lymph node metastasis(P=0.018), deeper invasion of tumor(P<0.001), vascular invasion(P<0.001), and lymphatic vessel invasion(P<0.001). For patients with hepatic metastasis(P=0.002) and lung metastasis(P=0.030), their contents of SDF-1α in peripheral blood was significantly higher than that without distant metastases. Logistic regression analysis revealed that invasion depth of tumor(P=0.027)was independent risk factor for SDF-1α content. Survival time of patients with higher content of SDF-1α was significantly shorter than that in patients with lower content of SDF-1α(P<0.001). Cox proportial hazard regression model analysis demonstrated that TNM stage(P=0.009), vascular invasion(P=0.002) and contents of SDF-1α in peripheral blood(P=0.001) were independent risk factors for patients survival.Conclusions: The increased contents of SDF-1α in peripheral blood might suggest the occurrences of lymph node metastasis, hepatic metastasis or lung metastasis and closely relate with metastasis and prognosis of gastric cancer.
