Objective: observing the changes of the mtDNA4834 bp and the Nrf-1 protein of the mimetic aging inner ear in the D-galactose-induced middle-aged rat. Methods: detecting the expression levels of mtDNA4834 bp of rats in the inner ears with real-time quantitative PCR, testing the expression levels of the NRF-1 in the rat inner ears with Western blot. Result: Due to the P<0.05 of the experimental group and control group, it is a statistical difference in the deletion mutation rates o f mtDNA4834 bp and expression levels of the NRF-1 in the inner ear. Conclusion: the damage of mitochondrial function is possibly increased by D-galactose in the inner ear of middle-aged rat, and D-galactose possibly activates the Nrf-1 mediated mitochondrial proliferation in the inner ear.
