Objective: 1.To explore the expression of NF-kBp65 and OPN in glandular epithelial cells of normal endometrium and ectopic endometriosis and eutopic endometrium, and their correlation;2.To analyse the change of the expression of NF-kBp65 and OPN in eutopic endometrial glandular epithelial cells of patients with endometriosis after OPN gene upregulation and downregulation and further to reveal the relationship between the two in endometriosis; 3. To study the effect on the invasion of endometrial epithelial cell after OPN gene up-regulation and down regulation.Methods 1. To collect endometriosis patients 35 cases as case group, Of which 18 cases of secretory phase, 17 cases of proliferative phase. To collect eutopic endometrium of patients for tubal sterilization 30 cases as control group, Of which 15 cases of secretory phase, 15 cases of hyperplasia in the same year.The immunohistochemical methods were used to detect the protein expression of NF- kappa Bp65 and OPN. 2. The glandular epithelial cells of endometriosis were primary isolated, cultured, OPN siRNA and plasmid interference endometrial glandular epithelial cells by RT-PCR and Western blot were used to detect the changes of the protein and mRNA expression of NF- kappa Bp65 and OPN.3. OPN and siRNA plasmid interference of endometrial epithelial cells by membrane assay of invasive change.Results 1.(1)Immunohistochemical results showed that: NF- kappa B p65, and OPN protein mainly expressed in endometrial epithelial cells cytoplasmic, a few in cells mesenchymal,and few in the vascular endothelial cells,and which the expression of the two in the eutopic and ectopic endometrium was significantly stronger in normal endometrium, and the expression of eutopic endometrium in the normal endometrium is more stronger;(2) The expressiom of NF- kappa B p65 protein in the proliferative and secretory phase endometrium, there was no difference, and was not statistically significant.The expression of OPN protein in three groups in the secretory phase was higher than that in the proliferative phase, at the same time,the expression of the two in eutopic endometriosis, ectopic endometrium was positively correlated. 2.The expression of NF- kappa Bp65, OPN mRNA and protein in the gland epithelial cells reduced and increased correspondly after OPN siRNA interference and OPN plasmid intervention,and the difference was statistically significant(P<0.05). 3. The invasion of gland epithelial cells in patients with endometrium reduced and increased correspondly after OPN siRNA interference and OPN plasmid intervention,and the difference was statistically significant(P<0.05).Conclusion 1. The expresssion of NF- kappa Bp65, OPN in eutopic and ectopic endometrium are high,and there was a significant positive correlation with the two. 2. OPN siRNA and OPN interference plasmid in eutopic endometrial epithelial cells, NFkappa Bp65, OPN expression was significantly decreased and increased, revealed OPN through NF- kappa B pathway and lead to the occurrence of endometriosis disease. 3. OPN siRNA and OPN plasmid interference in eutopic endometrial epithelial cells,the invasion of epithelial cells was significantly decreased and increased, It is suggested that OPN is a key factor to induce cell invasion.
