Objective: To investigate the expression of the mi R-210 and HIF-1? in the testis and epididymis of the varicocele rats. To analyze the relationship between mi R-210 and HIF-1? in testis and epididymis in each group.To master surgical modeling technology and varicocele ligation.To explore the significance of varicocele ligation.To explore the mechanism of varicocele and whether mi R-210 and HIF-1? can become a surgical indication or predict indicator of varicocele following-up.Method: 1.We choose 21 male Sprague-Dawl(SD) rats,and divide into control group(7),the experimental group(7) and surgical group(7) after a week.We operate on abdominal incision,narrow the left renal vein and ligature the vein between left spermatic vein and the left iliac vein under a microscope in experimental and operation group.The control group is exposed as same as the experimental group, but not narrow and ligature vein,closing the incision,and keeping 8 weeks.Then we measure spermatic vein diameter in control and experimental rats,and collect the left testis(weight testis) and epididymis,then part of them were fixed by formaldehyde and the others were frozened at refrigerator(-80 ?).We ligature left anesthesia on operation group rats,and feeding 4 weeks,then taking the same operation as the experimental rats. 2.We take a little of the testis or the head of epididymis tissue.Then the mic RNA extracted by mic RNA Extraction Kit is detected by real-time PCR.And the HIF-1? extracted by protein extraction kit is detected by Western Blot. 3. The relationship of mi R-210 or HIF-1? is analyzed between groups. We use the related analysis to analyse the association between mi R-210 and HIF-1? in groups.Results: 1. At last every group has 5 rats.The pathology shows the number and distribution of spermatogenic cells is worse, inflammatory, no sperm in the experimental group.It is better in the operation group.The diameter of the left spermatic vein in experimental group significantly enlarged than control group. The diameter of the left spermatic vein in operation group significantly diminished than experimental group. The diameter of the left spermatic vein in operation group enlarged than control group. All groups are statistically significant.2.The average weight of the left testis in the experimental group significantly decreased compared with the control.The average weight of the left testis in the operation group decreased compared with the control.The both groups are statistically significant. The average weight of the left testes in the operation group is not significantly changed compared with the experimental group.The group is not statistically significant. 3.The expression of mi R-210 and HIF-1? in the left testis and the epididymis of experimental group significantly increased compared with control group. The expression of mi R-210 and HIF-1? in the left testis and the epididymis of operation group is significantly decreased compared with experimental group.The two factors of both groups are statistically significant. The expression of mi R-210 and HIF-1? in the left testis and the epididymis of operation group is not significantly changed compared with control group.The group is not statistically significant. 4.The correlation of mi R-210 and HIF-1? in left testis and epididymis: Correlation analysis showed that they are positive correlation between the two factors.Conclusion:1.Testicular degeneration, weight loss and spermatic vein diameter enlargement is caused by varicocele. After ligation, testicular and epididymis damage is repaired, and spermatic vein diameter is also diminished. 2.The expression of mi R-210 and HIF-1? in the left testis of the varicocele rats was increased,and was decresed when ligation. The expression of mi R-210 and HIF-1? in the left epididymis of the varicocele rats was increased and was decresed when ligation too.The both factors in the groups are positive correlation. 3.The expression of mi R-210 in the left testis and epididymis is adjusted by varicocele.The expression of mi R-210 and HIF-1? is positive correlation, suggesting that they may have a relationship between the molecular pathways of varicocele.We predict that this may be one of the molecular mechanisms of testicular and epididymis damage. Perhaps the mi R-210 and HIF-1? will become an effective clinical indicator of varicocele on the impact of testicular and epididymis damage and varicocelectomy.
