Radix Astragalus,which is the dried root of Astragalus menmbranaceous(Fisch.)Bge.and Astragalus membranaceous(Fisch.)Bge.var.mongolicus(Bge.)Hsiao,has been widely used as a tonic in traditional Chinese medicine.It is sweet in taste and warm in property,and enters the lung and spleen meridians by tropism.Astragali has an effect of invigorating qi and consolidation of exterior and inducing diuresis for removing edema,and also detoxication and promoting granulation.Honey-fried Astragali is a product of raw astragalus mixed with honey by a traditional Chinese medicine processing method,which can strengthens the tonic,when compared to raw Astragali.Modern pharmacological studies have revealed that honey-fried Astragali can increase the immunity and enhanced phagocytic activity of macrophages.Currently,existing many reports for analyzing the changes of chemical constituents in raw Astragali and processed products had some problems,and there is still very few evaluation,the mechanism is not clear,conclusions are not uniform and so on.In addition,the role of honey in processing has not been scientifically demonstrated.Therefore,this study prepared Honey analogs-fried Astragali to reveal important ingredients in honey.There were four groups:Astragali group,fried-Astragali group,honey-fried Astragali group,honey analogs-fried Astragali group.A series of experiments including the change of chemical constituents,metabolic profiling,content of active ingredient,the biological activity and animal models of leucopenia of CTX were carried out in Astragali and processed products to compare the change of quality and explore the effect of honey-processing on the quality of Astragali.By setting the verification experiments,honey may play a role in Astragali from the processing to the extraction process.It is desirable to provide a reference for the mechanism to reveal honey-processed Astragali.The main contents were listed as follows:1.Honey analogs,which was prepared by the pharmaceutical excipients,was an alternative to traditional honey to processing.It had many advantages of safety,controllable quality and simple ingredients.From the perspective of chemistry and cell activity,it revealed that the major effective ingredients in honey were sugars during processing.2.HPLC/Q-TOF-MS was used to compare the metabolic profiling and the relative content of main components in Astragali and its processed products.Fingerprint analysis showed that it had little effect on the chemical composition,but large differences in the content of the chemical constituents.Metabolic profiling analysis showed that honey-fried Astragali and honey analogs-fried Astragali were a group and that raw Astragali and fried Astragali were divided into two groups,which showed that the addition of honey affected metabolic profiling and honey and analogs honey had similar effects.Semi-quantitative analysis showed that the content of acetylated products of glycosides in fried-processing significantly increased,while the content of aglycone decreased and glycosides increased in honey-fried Astragali.These results suggested that it may be due to high temperature in fried-processing,resulting in an increase in the content of acetylated products.Honey may be able to protect the glycoside so as not to be acetylated in honey-fried Astragali.3.HPLC-DAD-ELSD was used to compare the content of main components in Astragali and its processed products.The results showed that different processing methods had different effects on the content of chemical constituents.Glycosides remained basically unchanged in the water extract of fried-Astragali,while increased in honey-fried Astragali.Aglycone decreased significantly in the water extract of fried-Astragali,which also appeared in honey-fried Astragali.It showed that heating may cause the conversion of aglycone,and the honey was conducive to increased glycosides.Honey played an important role in the processing.4.From the point of the extraction rate,the stability and chemical transformation,experiments studied the effect of honey on extraction rate of total flavonoids and total saponins,thermal stability of pieces,water extract solutions,and saponins.The results showed that honey may promote the dissolution and transformation and protect the stability of some chemical compositions and so on.5.Animal models of leucopenia of CTX were carried out in Astragali and honey-fried Astragali to compare the change of pharmacodynamics by white blood cells,red blood cells,immune organs,body weight and other index parameters.Results showed that the Astragali and honey-fried Astragali antagozized CTX-induced leukopenia,atrophy of immune organs,increased the number of red blood cells and hemoglobin.It also improved immunity of mice.Immunological enhancement effect of honey-fried Astragali is better than Astragali. |
