Objective:To observe the effect of electroacupuncture at "Shenmen" 、"Sanyinjiao" of CRH pre processing model of insomnia rats sleep wake, emotional state and HP AA function effect, revealing electroacupuncture treatment of insomnia of central mechanisms and key neurotransmitter in.Methods:Healthy SD rats were weighed and the open field test selected experimental rats were suitable, and the qualified rats were randomly divided into blank group, sham injection group, CRH model group, CRH EA group. Take intracerebroventricular injection of drugs on insert only not to sham injection group, CRH the model group, CRH EA group rats to establish the model of insomnia take intracerebroventricular injection of CRH. After intracerebroventricular injection of 24 hour circadian locomotor activity and the open field test. After test of Electroacupuncture on CRH EA Shenmen, Sanyinjiao treatment, night 20:00 treatment begins,1 times/day for 4 days contining. After treatment for 24 hours a day on the spontaneous activity of the rats, the open field test. The behavioral test after experimental rat femoral artery blood 3~4ml, and immediately decapitated the thalams、brainstem, and rats central detection by ELISA methodperipheral plasma ACTH、CORT、5-HT and(thalamus、 brainstem) CRH、5-HT levels.Results:1.24 hour circadian locomotor activity:① After intracerebroventricular injection, Daytime activity:CRH model rats 24 hours circadian locomotor activity Daytime activity increased significantly compared with the blank group, CRH EA group rats 24 hours circadian locomotor activity Daytime activity increased significantly compared with the control group, were statistically significant (P< 0.05); CRH model group the rats 24 hours circadian locomotor activity Daytime activity significantly increased compared with sham injection group, CRH EA group rats 24 hours circadian locomotor activity and Daytime activity significantly increased compared to sham injection group, were statistically significant (P< 0.05); The night activity:sham injection group rats 24 hours circadian locomotor activity at night activity compared with the control group decreased slightly, with statistical significance (P< 0.05); CRH model rats 24 hours circadian locomotor activity night activity decreased significantly compared with the blank group, CRH EA group 24 hours a day Spontaneous night activities are active at night and blank group was significantly reduced compared has statistical significance (P< 0.05); CRH model rats 24 hours a day and night spontaneous nocturnal activity and sham injection group was significantly reduced compared, CRH EA group rats 24 hours a day and night spontaneous nocturnal activity and sham injection group was significantly reduced compared with statistical significance (P< 0.05);②After treatment:Daytime activity:CRH model rats day activity compared with the control group increased significantly, CRH EA group after the treatment of daytime activity compared with the blank group increased slightly, were statistically significant (P<0.05); CRH EA group after treatment during the daytime activity with the CRH model group was significantly reduced compared with statistical significance (P<0.05); The night activity:CRH model rats night activity compared with the control group decreased significantly, the difference was statistically significant (P<0.05); CRH model rats night activity and sham injection group decreased obviously, the difference is Statistical significance (P<0.05); CRH EA group compared with the CRH model group, the amount of night activity was significantly increased, the difference was statistically significant (P<0.05).2.Open field test:①Baseline:each group Open field test peripheral lattice score, central grid points, vertical scores are used single factor analysis of variance, the difference was not statistically significant, indicating that the baseline Open field test indexes balanced results, with the comparability.②The intracerebroventricular injectionafter:peripheral lattice score:CRH model group peripheral lattice score compared with the control group decreased significantly, CRH EA group peripheral lattice score compared with the control group decreased significantly, were statistically significant (P< 0.05); Statistical significance (P<0.05); ② The central lattice score:CRH model rats central square score compared with the control group increased significantly, CRH EA group after the treatment of the central lattice score compared with the control group increased slightly, were statistically significant (P<0.05); CRH model groud rats in central square and the scores of sham injection group significantly increased compared to a slight increase, the central lattice compared with sham injection group CRH EA group after treatment were statistically significant (P<0.05); CRH EA group rats after treatment with CRH score in central square of model group was significantly reduced compared with, with statistical significance (P<0.05); ③Vertical score:CRH model rats open field vertical score compared with the control group increased significantly, CRH EA group after the treatment of open field vertical score compared with the control group increased slightly, were statistically significant (P<0.05); CRH model group rats in the open vertical score significantly increased compared with sham injection group, CRH EA group after the treatment of open field vertical score and sham injection group compared to a slight increase, were statistically significant (P<0.05); CRH EA group after treatment was divided into vertical and Compared with the CRH model, the model was significantly reduced, with statistical significance (P<0.05).3. Plasma neurotransmitters 5-HT、ACTH、CORT quantity: ①CRH model group rats Peripheral Plasma 5-HT、ACH、CORT and blank group, sham injection group were significantly increased compared with the compared with statistical significance (P<0.05); ②CRH EA group rats after electroacupuncture therapy in Peripheral Plasma 5-HT、ACTH、CORT and CRH model group decreased significantly compared compared with statistical significance (P<0.05).4.CRH、5-HTof the central thalamus、brainstem: ①CRH model group rat thalamus、brainstem and neurotransmitter CRH in blank group、sham injection group were significantly increased、the difference was statistically significant (P<0.05); CRH EA group after treatment of thalamus、 brain stem neurotransmitter content decreased significantly compared to CRH with the CRH model group, the difference was statistically significant (P<0.05); (2) CRH model group rat thalamus、brain stem neurotransmitter content of 5-HT compared with the blank group、sham injection group were slightly reduced, the differences were statistically significant (P<0.05); CRH EA group after treatment of thalamus、brain stem neurotransmitter the content of 5-HT increased slightly compared with CRH group, the difference was statistically significant (P<0.05).Conclusion:Intracerebroventricular injection of CRH on rats, can cause insomnia model; the hypothalamic paraventricular nucleus CRH is HPAA electroacupuncture regulation function of rat model of insomnia, the key material for improving sleep state; hypothalamus the rat model of insomnia pituitary adrenal axis increased excitability; the Electroacupuncture of "Shenmen" 、 "Sanyinjiao" can be hindered by the hypothalamic pituitary adrenal axis hyperactivity to adjust insomnia.
