Curcuma phaeocaulis Val.is one of the main source of Turmeric.According to the Chinese Pharmacopoeia of 2015,Turmeric can promote the circulation of qi,relieve pain,activate blood stagnation and expel blood stasis.The doctor of TCM usually use Curcuma phaeocaulis VaL processed with vinegar for "Effect-enhancing",which is not clear from the modern research.ObjectiveThis thesis aims to unveil the scientific connotation of Curcuma phaeocaulis Val.processed with vinegar for "Effect-enhancing",including the change on both sides of the component and effective,based on protein-protein interaction network to predict the targetsMethod and Result1.The raw product of Curcuma phaeocaulis Val.was processed with 9° vinegar,purified water and 9%acetic acid aqueous solution to be vinegar product blank control and positive control,meanwhile measuring the pH at different time of each group.The result showed that the pH of vinegar product and positive control increase from 3.8 to 4.8,blank control decreased from 7.1 to 6.3.Then each group was decocted and also measured the pH at different time.The result showed the pH of vinegar product and positive control was below 6,lower than blank control and raw product.2.A HPLC method was developed for determination of curcuminoids(curcumin,demethoxycurcumin,and bisdemethoxycurcumin)and sesquiterpenoids(β-elemene,curcurmol,germacron,and curdione)at the same time.Then each group were determinated.The result showed that the content of sesquiterpenoids were higher compared to the content of curcuminoids,while sesquiterpenoids decreased more than curcuminoids in processed product;The contents of curcuminoids in processed product and positive control decreased less than that in blank control group.3.Con-A induced hepatitis mice were adopted to compare the anti-hepatitis effective of Curcuma phaeocaulis Val and its vinegar product.The raw product and the processed product group were given the ethanol extraction(1.3g/kg),the positive control group were given bifendate(200mg/kg);the referent groups were given curcumin(200mg/kg)and demethoxycurcumin(200mg/kg)and the negative control group and normal group were given blank solution.All the groups were i.g.for 7 days,then injected Con-A.The liver index,spleen index,ALT,AST,NO,T-SOD,MDA,LDH were determinated.The result showed that each dosing group have improvements compared to model group;the effective of processed product is more close to the bifendate,better than raw products.4.The protein interaction network(PIN)was employed to predict the targets of Curcuma phaeocaulis Val.on anti-hepatitis.A database of the components in Curcuma phaeocaulis Val.was developed.Then the information of the componets’targets were obtained from the STITCH 4.0 and ChEMBL databases.The protein interaction network of Curcuma phaeocaulis Val.was constructed including 723 nodes and 2726 edges by Cytoscape.According to the topological analysis,the network is stable and reliable.Based on module analysis,there are 25 functional modules including the immune regulation process and TGF-β signaling pathways process and so on.It is found that curcumin and bisdemethoxyciucumin can involve in signaling pathways process,regulating TGFβ1,TGFβR1,TGFβR2,SMAD3,SMAD4,ZFYVE9 to anti-hepatitis.5.Western Blot and qPCR were used to detect the level of gene TGFβ1,Smad7 and Smad2/3 expression and protein contents in the liver of each group,it was found that the raw product and the processed product can regulate the TGF-β signal pathway.Curcumin and bisdemethoxycurcumin are the main active components.ConclusionThe scientific connotation of the vinegar processing on Curcuma phaeocaulis Val.is "protecting the insufficiencies,leaving out the surplus".which reflects in that after vinegar process,the contents of sesquiterpenoids are greatly reduced by the heat,while the curcuminoids are effectively protected by vinegar,mainly for the acetic acid.The curcuminoids are the main active components of Curuma phaeocaulis Val.to anti-hepatitis,for the ability to regulate the TGF-β signal pathway.This research enriched the scientific connotation of effect-enhancing to a certain extent,and provide a new clue for the processing mechanism research.
