Objectives:1.Through the establishment of rabbit ear scar hyperplasia model, it is proved that the silicone oil urea dexamethasone cream has the effect of inhibiting the proliferation and collagen synthesis of rabbit ear fibroblasts to inhibit the proliferation of hypertrophic scar.2.To observe the effect of silicone oil urea cream on Thl/Th2 related cytokines and TGF-beta 1/Smad3 signal transduction pathway in the model animal scar tissue, and to explore the possible mechanism of silicone oil urea dexamethasone cream in the treatment of hypertrophic scar.Methods:After rabbit ear hypertrophic scar model was established,21 days of successful modeling were randomly divided into model group, positive control group, experimental group, Chinese medicine group, daily to be 2 times the applicator used weekly, vernier caliper to measure the rabbit ear scar thickness. After 14 days and 28 days, the rabbit ear scar tissue was taken for pathological examination. The cases of fibroblast and collagen were observed, and the pathological changes of the scar tissue were observed. ELISA method was used to determine the content of Thl/Th2 related cytokines IFN- and IL-4 protein, and q-PCR method was used to detect the expression of TGF- beta 1 and Smad3 protein in scar tissue.Results:1.Compared with the model group, silicone oil urea dexamethasone cream (high, medium. low) concentration in the group and Chinese medicine group proliferation index decreased significantly (p< 0.05). The results showed that the contents of fibroblasts and collagen were significantly decreased (p<0.05), and the arrangement of collagen fibers was improved in the scar tissue of rabbit ears.2. The model group compared with normal group, IFN-γ protein content decreased, IL-4 protein content increased (p< 0.05); silicone oil urea dexamethasone cream (high, medium, low) concentration group and Chinese medicine group compared to model group,IFN-γ protein content increased, IL-4 protein content reduce (p< 0.05).3. The model group compared with normal group, TGF-beta 1, smad3 gene expression were significantly increased (p< 0.05); silicone oil urea dexamethasone cream (high, medium, low) concentration group and Chinese medicine group compared to model group,TGF -beta 1 and smad3 protein gene expression were decreased (p< 0.05).Conclusions:1. The effect of silicone oil urea dexamethasone cream on the proliferation of hypertrophic scar in rabbit ears was obvious.2. Th1/Th2 imbalance is one of the mechanisms of hypertrophic scar formation in rabbit ears. Silicone oil urea dexamethasone cream and Sodium ferulate-sinomenine ointment throng reversing the Th1/Th2 imbalance, thereby inhibiting the proliferation of rabbit ear scar.3.TGF-beta 1/smad3 signaling pathway is one of the mechanisms of hypertrophic scar in rabbit ears. silicone oil urea dexamethasone cream and Sodium ferulate-sinomenine ointment can inhibit the proliferation of rabbit ear scar by inhibiting the TGF-beta 1/smad3 signaling pathway.4.Thl/Th2 related cytokines have the role of regulating the TGF-beta 1/smad3 signaling pathway, and participate in the formation of rabbit ear scar formation mechanism. silicone oil urea dexamethasone cream and Sodium ferulate-sinomenine ointment can inhibit the proliferation of rabbit ear scar by inhibiting the transduction of Thl/Th2 related cytokines in TGF-beta 1/smad3 signal.
