| Dark tea is one of the six major tea categoric many previous research showed that dark tea has the functions of lipid-lowering and in some extend hypoglycemic effect. Fuzhuan brick tea, a type of dark tea, which contains a special component named eurotium in it.In this study, Based on the extract from Fuzhuan brick tea, the simulation of most the reasons triggering diabetes Ⅱ, and mice models carrying diabetes Ⅱ deduced by high glucose feeding and streptozotocin(STZ), it can be proved that the brick tea has a significant effect on controlling the glucose and lipid metabolism disorder with the dose-effect relationship, after referring to analysis of Animal Morphology Symptoms of phenotypic, physiological, biochemical index and the gene expression. The test results are as follows:1.The intervention effect of diabetes Ⅱ mice by Fuzhuan brick tea.According to the analysis of Animal Morphology and Symptoms, the conclusion can be drawn from the brick tea’s interference on the mice with diabetes Ⅱ.It shows that the brick tea can not only significantly reduce the blood sugar levels in the animal’s body with HFT dropping to 37%(P<0.01),but increase the endurance to glucose load with AUC in HFT decreasing by 30%(P<0.05),The body’s insulin resistance index rising by 47%(P< 0.01), and insulin sensitivity level by 38%(P< 0.01).What’s better is that can be capable to improve the water consumption of those diabetes mice and control their weight-loss, along with dry cages, diuresis is under control and limbs ulcers of mice being alleviated.2.Fuzhuan brick tea’s function on improving redox imbalance of diabetes Ⅱ mice.The physiological and biochemical index indicates that the tea can reduce lipid metabolism and blood lipid levels for the TC, TG, HDL decreased in HFT group and the AST,ALT (P<0.05) estimating the liver dysfunction dropped. MFT and HFT significantly increased SOD (17%, P<0.01), GSH-px (28%, P<0.01) and CAT (26%, P<0.01) activity, and decreased XOD activity (19%, P<0.05) and MDA level (37%, P<0.01). Therefore,the tea is conducive to improving the disorder of lipid metabolism, regulating peroxidation in mice, preventing and mitigating for diabetes caused by acute and chronic complications so as to protect the liver.3.The mechanism of the tea’s improving diabetes Ⅱ through PI3K/Akt signal path.The QRT-PCR analysis on gene closely related to glucolipid metabolism shows that, the tea can significantly increase and eight fat splitting and digestive GLUT2 gene PPAR alpha and sugar transport important genes and gene expression of GLUT4 quantity (PPAR alpha: 1.886, P< 0.05; GLUT2:1.678, P< 0.05; GLUT4:2.430, P< 0.05), while the energy supply AMPK and regulation of lipid metabolism related genes ppar-gamma regulation to the normal level (ppar-gamma AMPK-alpha:1.586:1.583), but there was no significant difference (P> 0.05). The tea extract significantly enhances the body of insulin secretion and utilization and promote glucose transporter and lipid metabolism in the liver, sugar metabolic disorder to improve the condition of diabetes mice. |
