Scutellaria barbata herba is a traditional Chinese medicinal herb which is commonly utilized. From the perspective of traditional Chinese medicine, it has the effects of heat-clearing and detoxifying. Modern pharmacological studies reveal that Scutellariae barbatae herba contains a variety of medicinally active substances which have pharmacological effects such as anti-tumor, antibacterial, antioxidation, anti-inflammation, antipyretic effect and immunoregulation. This dissertation focuses on Scutellaria barbata herba. In order to provide a reliable basis for quality control and modern preparation of active constituents from Scutellaria barbata herba, the determination method of 4 flavonoids and the fingerprint of Scutellaria barbata herba were established by HPLC, and the integrated extraction-adsorption process was developed and applied to extract and purify total flavonoids from Scutellariae barbatae herba. The content includes the following parts.1. Establishing the HPLC analysis method of 4 flavonoids in Scutellaria barbata herbaThe HPLC determination method of scutellarin, scuellarein, luteolin and apigenin was established by investigating the pretreatment methods of the sample and optimizing the chromatographic conditions. Methodological study showed the method was highly specific and had good precision and accuracy. It was confirmed that the differences of the contents of 4 flavonoids were large between different batches of Scutellaria barbata herba.2. Establishing the HPLC fingerprint of Scutellaria barbata herbaThe fingerprint of Scutellaria barbata herba was established by HPLC. Methodological study showed the method was stable and reliable, which could be utilized in quality control.10 batches of Scutellaria barbata herba were analyzed by the method with 12 common peaks separated. "Similarity Evaluation System for Chromatographic Fingerprint of TCM" was used to evaluate the similarities. It turned out that the similarities of 9 batches were over 0.90 and the quality differences were not significant between batches.3. Preparing total flavonoids from Scutellariae barbatae herba by integrated extraction-adsorption processIntegrated extraction-adsorption process was developed and applied to extract and purify total flavonoids from Scutellariae barbatae herba. Screening of macroporous resins, single factor experiments, an experimental design based on Box-Behnken design (BBD) and purification conditions study were carried out to optimize the integrated process. DM130 resin was chosen as adsorbent and the optimal extraction-adsorption conditions were determined as pH of extraction solvent 2, time 9 h, liquid/solid ratio 20:1 mL/g and flow rate 7 BV/h.5 BV of 70% ethanol aqueous solution was selected as elution solvent. Through this procedure, the recovery rate of the total flavonoids was 46.70 mg/g and the purity was 69.03%.
