Gastric cancer is a kind of digestive tract malignant tumor,are distributed throughout the world.The incidence is higher in east Asia(including China,Japan and South Korea,etc.)than in Europe and north Amercia.Gastric cancer early diagnosis rate is low in our country,most patient is advanced.The recurrence and metastasis of malignant tumor are major causes of death in patients,but the comprehensive assessment system of risk of recurrence and metastasis of gastric cancer patients is not perfect,so we need to find more effective indicators to improve it.GGT1(gamma glutamyl transpeptidase 1)play a key role in glutathione metabolism and in the transformation of leukotriene C4 to D4.GGT1 can be used as tumor markers in certain types of cancer.There is no relevant report about GGT1 in gastric cancer research.Tissue chip technology can analysis of a large number of clinical specimens at the same time,possesses the advantages of small size,large amount of information,also has the advantages such as high efficiency,rapid and low loss.Its application in molecular pathology of tumors and clinical pathological diagnosis is becoming more and more widely.AimDetect GGT1 protein expression in gastric cancer tissue,evaluation its clinical significance and find effective predictors of gastric cancer.MethodThe paraffin specimens of gastric cancer patients were made into tissue microarray,detected GGT1 protein expression by immunohistochemical technique,and its relationship with clinical pathological factors and its influence on the prognosis of patients with gastric cancer were analyzed.Result1.The paraffin specimens of 472 cases of patients with gastric cancer were made into tissue microarray,eventually 322 organization point of them can be analysised.2.GGT1 was positive expressed in the cytoplasm.GGT1 expression is associated with lymph node metastases(P = 0.017),is associated with gender(P = 0.015),and associated with the expression of Her2 expression(P=0.003).3.High expression GGT1 obvious influence on the overall survival of patients with gastric cancer(log-rank p<0.01).4.Multiariable Cox risk model showed that GGT1 can better predict he poor prognosis of gastric cancer patients,such as advanced(III/IV,HR = 1.63;95%CI,1.10 to 2.39),low differentiation(HR = 1.98;95%CI,1.31 to 2.96),undifferentiated carcinoma(HR = 1.65;95%CI,1.04 to 2.56),Mucinous&Signet Ring Cell carcinoma(HR=3.81;95%CI lL53-8.25)and Her2 positive(HR = 2.42;95%CI1.35-2.42)Conclusion1.It is feasible to detect clinical tissue samples with tissue microarray technique,also it is fast,convenient,economic and accurate.2.GGT1 expression is associated with lymph node metastases,the expression of Her2 expression and gender.3.The high expression of GGT1 obvious influence on the overall survival of patients with gastric cancer.4.GGT1 can be used as independent factor of adverse outcomes for patients with gastric cancer.GGT1 can better predict he poor prognosis of gastric cancer patients,such as advanced,low differentiation,undifferentiated carcinoma,Mucinous&Signet Ring Cell carcinoma.
