| microRNAs are a class of small,non-encoding endogenous RNAs in plants and animals,and regulate expression of 30%genes in the human genome.Many evidences indicate that change of miRNAs expression is one of common features of tumor,and miRNAs in tumor cells involve in regulation of cell cycle,migration,invasion and apoptosis.miRNAs can be used as oncogenes or tumor suppressor genes,in addition to cell metastasis.miRNAs can be also used as molecular markers for early diagnosis of cancer.miR-490-3p is down-regulated and may act as a tumor inhibitor in many tumors.In addition,miR-490-3p shows a significant change in heart diseases and would be closely related to heart diseases.However,specific roles of miR-490-3p are not well understood in cancer and heart diseases.This study aims to investigate the effect of miR-490-3p on tumor cell proliferation,and explore the role of miR-490-3p in cardial development and diseases.The expression of miR-490-3p in serum of peripheral blood in myocardial in-farction patients and normal controls were detected by RT-qPCR,and showed an up-regulated pattern in myocardial infarction patients,suggesting that miR-490-3p can act as a potential diagnostic marker for myocardial infarction.Glucose uptake and lactic acid production were reduced in miR-490-3p over-expression H9C2 cells,In addition,in order to investigate effect of miR-490-3p on tumor proliferation and metabolism,miR-490-3p over-expression cells were obtained by transfection,the expression levels of downstream genes were detected using Western blotting and RT-qPCR.Concentra-tions of glucose and lactic acid were analysed using glucose detection kit and lactic acid detection kit.Cell proliferation was measured by cell counting,the ability of migration was detected by wound healing assay,and cell cycle was assayed by flow cytometry.These analyses showed that miR-490-3p inhibited expression levels of hnRNPA1 pro-tein and mRNA,promoted expression of PKM1,and decreased ratio of PKM2/PKM1 expression.These changes thus reduced glucose uptake and lactic acid production,inhi-bited cell growth and migration,arrested HeLa cells in G2/M phase and 786-0 cells in Gl/GO phase,and inhibited EMT of tumor cells.These results suggested that miR-490-3p can be used as a potential therapeutic target and a molecular marker for cancer prediction. |
