Background and Objective:Guided tissue regeneration(GTR)or guided bone regeneration(GBR)is a procedure,which has been widely used in periodontal regenerative medicine and perimplant bone regeneration.New generation of barrier membranes has been developed,which not only act as barriers but also as delivery devices to release specific growth factors.BMP-2 provides the good bone regeneration potentials evidenced by animal models and human trials,and basic FGF.also called as FGF-2,is another kind of well-known growth factor regulating cell proliferation,migration and differentiation.However,whether loading BMP-2 or bFGF onto ADM membranes can potentialize bone regeneration ability and whether there is any different effect of BMP-2-and bFGF-ADM membranes on bone regeneration process remain to be explored.This study evaluated differential bone regeneration process stimulated by bFGF and BMP-2 loaded into acellular dermal matrix membrane(ADM).Materials&Methods:0.5 ml 200 ng/ml bFGF or 800 ng/ml BMP-2 was loaded onto ADM membranes(10×8×1 mm)by soakinog.incubated at 4℃ overnight and then freeze-dried at-60℃.Critical-size rat calvarial defects were created and covered by ADM alone.ADM+bFGF(100 ng),ADM+BMP-2(400 ng)or blank control.Animals were sacrificed at scheduled time,then number of MSCs,osteogenisis and new bone volume were observed by immunohistochemistry,immunofluorescence,HE staining and KMicro-CT respectively.Results:ADM after freeze-drying treatment could sustainably release drug for about 10 days,although an initial burst release was observed.The in vivo experiment demonstrated 2-fold more CD34-/CD90+ MSCs in group of ADM loaded with bFGF treatment than in any other treatment groups at 2 weeks.However,there were similar amount of new bone formation and expression of OPN in newly-formed bone tissue in groups of ADM loaded with bFGF and BMP-2 at 8 weeks,which were more than those in ADM alone and blank control.Conclusion:Although both BMP-2 and bFGF can improve bone and periodontal regeneration,their effect mechanisms differ.bFGF loaded into ADM membrane enhances bone regeneration similar to BMP-2 by early recruition.sternness maintenance and the persistence for osteogenic differentiation of BMMSCs.Therefore.bFGF and BMP-2 all are suitable candidates for growth factors loaded on GTR membranes to reinforce bone regeneration.First release of bFGF and then BMP-2,is expected to possess better bone regeneration efficacy.
