| Monascus fermented products have been universally used in food and pharmacy-eutical industry in China.It contains a variety of secondary metabolites,such as r-ed pigments,a series of monacolins and so on.Monascus pigments used primarily as a natural colorant and monacolins as hypolipidemic active components,is an in-creasing concern in China.This paper mainly focus on the research of Monascus T-UST001 fermented production,after removing of Monacolin K,the effects of remain nnned Monacolin K Free(MKF)components in TUST001 fermented products on c-holesterol lowering mechanism in HepG2 cells were investigated.The proliferation inhibitory effects of ethanol extractions from fermentation pro-ducts using different varieties of rice as substrate on HepG2 were different.The pr-oliferation inhibitory effects of ethanol extractions of fermented products are better than Monacolin K standard alone.Comparing the inhibition effect of HepG2 cell pr-oliferation incubated with ethanol extractions of fermented product after sterilization or non-sterilization,we found that high temperature decreased the inhibition effect of HepG2 cell proliferation in fermented products.The different concentration of etha-nol extract of fermented products by TUST001 were cultured with HepG2 cells,Th-e cholesterol metabolism related genes were determined after 48h of incubation,the transcription expression of HMGCR,SREBP-2,LDLR,CYP7A1 and LXR-a increase ed slightly.In order to remove the Monacolin K from TUST001,high speed counter curre-nt chromatography and high performance liquid chromatography were employed,we-got three components:MKA,MKL and MKF with high speed counter current chro-matography,six components were acquired after separated with high performance li-quid chromatography.These components were acting with HepG2 cells respectively;the results showed that each component was able to inhibit HepG2 cell proliferation.Through elevating cholesterol concentration in cell with cholesterol quantitation kits,we found that dealing with MKF,the cholesterol concentration in HepG2 cells decr-eased 19.7%.Besides Monacolin K,there are other bioactive constituents in Monas-cus fermented production responsible for the cholesterol-lowering activity.Finally,we detected gene transcriptional expression concerning of cholesterol m-etabolism in HepG2 cells incubated with MKF.The transcriptional expression of H-MGCR and LXR-a had no change,the expression of SREBP-2,CYP7A1 in transcr-iptional level increased,and the transcriptional level of LDLR decreased. |
