Background and ObjectiveHepcidin has been known as a kind of polypeptide in the past years that is synthesized and secreted by the liver,playing an important role in the progress of iron metabolism.In recent years,the studys about the relationship of Hepcidin and the malignant tumors show that cancers always goes with abnormal serum Hepcidin levels,which not only implys that Hepcidin plays a central role in the progress of iron metabolism of cancer cells,but also suggests that there is some correlation between Hepcidin and the generation and progress of tumors.At present,the studys about the relationship of Hepcidin and the malignant tumors always focuse on the correlation between Hepcidin and cancer-related anemia.There is no research data on serum Hepcidin and its relationship with iron metabolism in children with acute leukemia at home.In this study,we tested the serum levels of Hepcidin,IL-6,transferrin(TRF)andferritin(FN)in children with acute leukemia by enzyme-linked immunosorbent assay(ELISA).We explored the most possible mechanism about the abnormal serum levels of Hepcidin in children with acute leukemia and the correlation between the Hepcidin and iron metabolism by analyzing the relevance among them.MethodsThe 63 cases of children with acute leukemia who were diagnosed from the pediatrics hematology ward of the first affiliated hospital of ZhengZhou University from November 2014 to November 2015 were selected as the object of the study,and they all received a follow-up visit until September 30,2016.Then,We divided them into two groups “the remission group and the non-remission group”.A group of 30 healthy children was used as a control.Collect the blood of the healthy children and the AL children before the treatment and after the treatment.The Hepcidin,IL-6 in serum were tested by enzyme-linked immunosorbent assay(ELISA).On the other hand,we also tested the level of TRF,FN in serum.Compare the expression differences of Hepcidin,IL-6,TRF and FN among every group.In the end,we analyzed the correlation between Hepcidin and IL-6,the correlation between Hepcidin and the iron metabolism indexes TRF,FN.Results1.The general situation of the inclusion of the objectThe difference about gender between AL group and the control group was not statistically significant by χ2 test(χ2=0.031,P=0.860).The difference about age between AL group and the control group was not statistically significant by independent sample t-test analysis(t=0.332,P=0.756).2.Expression levels of Hepcidin,IL-6,TRF,FN in serum in AL group and the control groupThe expression levels of Hepcidin,IL-6 and FN in serum of children with acuteleukemia in the initial treatment group were higher than the control group,and the differences between them were all statistically significant(P<0.001).The expression level of TRF in serum of children with acute leukemia in the initial treatment group was lower than the control group,and the difference between them was statistically significant(P<0.001).3.Expression levels of Hepcidin,IL-6,TRF,FN in serum in AL group before the treatmen and the afterThe expression levels of Hepcidin,IL-6 and FN in serum of children before the treatment who achieved remission were higher than the group after the treatment,and the differences between them were all statistically significant(P < 0.001).The expression level of TRF in serum of children before the treatment who achieved remission was lower than the group after the treatment,and the difference between them was statistically significant(P<0.001).The expression levels of Hepcidin,IL-6,FN and TRF in serum of children before the treatment who achieved non-remission were not significantly different from the group after the treatment(P>0.05).4.Expression levels of Hepcidin,IL-6,TRF,FN in serum in the remission group and the non-remission groupThe expression levels of Hepcidin,IL-6 and FN in serum of children in the non-remission group were higher than the group with remission no matter before and after the treatment,and the differences between them were all statistically significant(P < 0.001).The expression level of TRF in serum of children in the non-remission group was lower than the group with remission no matter before and after the treatment,and the differences between them were all statistically significant(P<0.001).5.The relationship between Hepcidin and IL-6 in serum;The relationship between Hepcidin and TRF,FN in serumIn the initial treatment group,the expression level of Hepcidin positively correlated with the expression level of IL-6(r=0.866,P<0.001).The expression level of TRF inversely correlated with the expression level of Hepcidin(r=-0.834,P<0.001).The expression level of FN positively correlated with the expression level of Hepcidin(r=0.750,P<0.001).Conclusions1.The expression level of Hepcidin increased significantly in children with acute leukemia and there is some disorder of iron metabolism in children with acute leukemia.2.The expression levels of Hepcidin,IL-6,TRF and FN in children with acute leukemia were different from those in healthy children,which suggests these indexes could be used as the diagnostic indicator with acute leukemia.The expression levels of Hepcidin,IL-6 and FN and TRF after the treatment were significantly different from the before in the remission group,and there are some significant differences about Hepcidin,IL-6,TRF and FN between the remission group and the non-remission group,which implys these indexes could be used to judge the effect of treatment.3.In the children with acute leukemia,the expression level of Hepcidin positively correlated with the expression level of IL-6 in serum,which reminders that IL-6 is one reason causing the higher serum level of Hepcidin.The expression level of TRF inversely correlating with the expression level of Hepcidin in serum,and the expression level of FN positively correlating with the expression level of Hepcidin in serum,suggests that Hepcidin participates in the disorder of iron metabolism in children with acute leukemia.between Hepcidin and TRF,FN in
