Objective: To observe the effect of Digeda-4 on the intrahepatic cholestasis of rats induced by naphthalene isothiocyanate(ANIT),to explore its pharmacological action and mechanism,and to provide scientific basis for clinical application。Method:Sixty SD rats were randomly divided into normal group,model group,ursodeoxycholic acid group(0.18 mg / kg)and ground weight-4 low(0.36 mg / kg),medium(0.72 mg / kg)(1.44 mg / kg)dose group,respectively,for 15 days,given the first 13 days of administration of ANIT(40 mg / kg)replication of cholestatic rat model.After 48 hours,the rats were anesthetized with chloral hydrate,and the serum was collected by centrifugation and centrifugation for 10 min.ALT,AST,ALP and TBIL were detected.The liver tissue was fixed from the same part of the liver.Sixty SD rats were randomly divided into normal group,model group,ursodeoxycholic acid group(0.18 mg / kg)and ground weight-4 low(0.36 mg / kg),medium(0.72 mg / kg)(1.44 mg / kg)dose group,respectively,for 15 days,given the first 13 days of administration of ANIT(40 mg / kg)replication of cholestatic rat model.After 48 hours,the rats were anesthetized with chloral hydrate and the levels of serum TNF-α,MDA,SOD,IL-6,GSH-PX and IL-10 were measured by centrifugation.Results:Digeda-4 can significantly reduce the AST,ALT,ALP and TBIL activity of ANIT-induced rat cholestasis rat model,and can effectively relieve the pathological changes of liver tissue.Digeda-4 Low dose,middle dose and high dose could significantly decrease the levels of serum TNF-α and IL-6,which were significantly different(P <0.01 or P <0.05).Digeda-4 low dose,medium dose,high dose can significantly increase the level of IL-10,there are significant differences(P <0.01 or P <0.05).Digeda-4 medium dose and high dose could significantly decrease the level of MDA in liver tissue.The low dose,middle dose and high dose could increase the level of SOD in serum and liver tissue,but there was no statistical significance(P> 0.05).Digeda-4 low dose,medium dose and high dose could significantly increase the GSH-PX level in serum and liver(P <0.01 or P <0.05).Conclusion: Digeda-4 can be through the inflammatory response and inhibition of oxidative stress this mechanism to treat intrahepatic cholestasis,improve the ANIT-induced intrahepatic cholestasis rat model of biochemical indicators,play a role in liver protection.
