| [Background and Objective]Non-alcoholic fatty liver disease(NAFLD)is a common clinic pathological condition characterized by significant lipid deposition in the hepatocytes of the liver parenchyma and persistent abnormalities in liver enzyme.According to the epidemiological investigation and study,non-alcoholic fatty liver disease is closely related to obesity.Immunoglobulin E(IgE)is well known for its role in allergic disease and participates in the process of parasitic infection.IgE high affinity receptor FcεRl is one of the IgE receptors.Yun-Jung Lee’s research found that,plasma IgE levels correlated inversely with body weight,body mass index,and body fat mass among a population of randomly selected obese women.FcεR1 deficiency in diet-induced obesity(DIO)mice increased food intake,reduced energy expenditure,and increased body weight gain.The aim of this investigation was to evaluate the efficacy of IgE high affinity receptor in the development of non-alcoholic fatty liver disease through the experimental non-alcoholic fatty liver disease model induced by high fat diet.[Methods]1.Model building and index detection:to induce NAFLD,both WT and FcεR1-/-mice from each group(n=10,male)were fed with a high fat diet for 12 weeks.Body weight was measured at a fixed time every week.All mice were sacrificed after 12 week treatment.Serum and liver were harvested to examine liver relevant parameters.2.Insulin tolerance test:mice were fasted for 6 hours after 10 weeks’ feeding and given 0.66IU/kg insulin through intraperitoneal injection.At the same time,blood glucose measured was at 0,30,60,90,120,150min by a glucose meter.3.Glucose tolerance test:mice were fasted for 12 hours after 11 weeks’ feeding and given 2g/kg glucose liquid through intraperitoneal injection.At the same time,blood glucose was measured at 0,30,60,90,120,150min by a glucose meter.[Results]1.Body weight and liver/body ratio:compared with WT mice,the growth of body weight and liver/body ratio in FcεR1-/-mice were significantly increased.2.Liver function and fibrosis deposition in the liver:compared with WT mice,serum alanine aminotransferase,aspartate aminotransferase levels and type Ⅲcollagen in the liver were significantly increased in FcεR1-/-mice,while type Ⅰcollagen in the liver exhibited no obvious difference.As revealed by HE staining and masson staining,the liver tissue has suffered damage and the fibrosis deposition was increased.3.Lipid metabolism:compared with WT mice,serum triglycerides,total cholesterol levels and triglycerides in the liver tissue were significantly increased in FcεR1-/-mice.As revealed by oil red O staining,the lipid deposition in the liver was increased.4.Glucose and insulin tolerance test:there was no obvious difference of glucose and insulin tolerance test in two groups.[Conclusions]Deficiency of IgE high affinity receptor FcεR1 in high-fat induced non-alcoholic fatty liver disease mice aggravated the symptoms of non-alcoholic fatty liver disease,increased body weight gain and increased lipid deposition in the liver.All results indicated that IgE high affinity receptor FcεR1 could regulate lipid metabolism and protect high-fat diet induced non-alcoholic fatty liver disease in mice. |
