Nano drug delivery system is a hot research field,especially for cancer research.Poly(glutamic acid)(PGA)is one of the most recognized medical polymer materials in the world.Poly glutamine(PGG)was optimized on the basis of PGA,and glutamic acid was added on the basis of glutamic acid.This provides more free carboxyl groups for polymers,greatly increasing the likelihood of PGA.This paper introduced a cosolvent in glutamic acid PGG phenylalanine ethyl branched,the free by chemical bond and carboxyl groups on the PGG connected to both the formation of a hydrophilic polymer and a free carboxyl group,hydrophobic phenylalanine.The polymer material obtained by physical encapsulation and paclitaxel nanoparticle formation,by NMR,dynamic light scattering,high performance liquid chromatography,scanning electron microscopy and the corresponding methods were used to characterize hydrogen displacement,particle size distribution,drug loading,projection type map,different pH environment nanoparticles the ability to release data on polymer materials PGG-PAE and PGG-PAE/PTX nanoparticles.The experimental results show that the average size of the prepared PGG-PAE/PTX is 70.7nm,which can meet the EPR effect,and can have the physical targeting effect of tumor.Dispersion of PDI is 0.073,very small,indicating that the size of PGG-PAE/PTX particles is almost the same,more uniform.The Zeta potential results also show that the nanoparticles are very stable and difficult to accumulate.Transmission electron microscopy showed that the nanoparticles were self-assembled into spheres,and the particle size corresponded to the DLS particle size map.It can be seen from the solubility test that the solubility of PGG-PAE to PTX is increased by 105 orders of magnitude,which is a good solution to the problem of water solubility of PTX,which can effectively improve the efficacy of PTX.The results of stability test showed that the particle size of nanoparticles did not change in two weeks,indicating that the nanoparticles were very stable,which also provided strong support for the physical and chemical properties of nanoparticles.The results showed that PGG-PAE/PTX was higher than PGG-PTX,indicating that the amount of physical release was higher than that of chemical connection.According to the data of drug release,the release rate of paclitaxel was two times more than that of chemical bond,which was more than 43%.These provide a good guarantee for the application of PGG-PAE/PTX.It can be seen from the test results that the soluble polymer nano material has opened up a very good application prospect for paclitaxel,and laid a solid foundation for the better application of paclitaxel in clinic.
