| Objective:NLRP3 inflammasome signaling pathway can regulate the secretion and maturation of cytokines such as IL-1β,and its roles in periodontal disease are important.The mechanism of porphyromonas gingivalis involved in periodontitis is more complicated.The present study aims to observe the expression of IL-1β,NLRP3,ASC,Caspasel in different types of gingival tissue and different treated THP-1 cells,and to explore the roles of this signaling pathway in chronic periodontitis.Methods:CCK8 kit was adopted to test the viability of THP-1 cells;and the IL-1β level was tested by Elisa kit;real-time PCR measures the relative gene expression of NLRP3,ASC,Caspaseland IL-1β in gingival tissue and different treated THP-1 cells.Western-Blot method was used to test the protein levels of the above targets.Results:THP-1 cells viability is related to the stimulating concentration and time lengthy,and the time effect is mainly reflected in the time points of 48h and 72h.IL-1βlevel in cell suspension is determined by the stimulation and concentration of P.gingivalis.The mRNA expressions of IL-1β and Caspasel were elevated along with the increasing time and concentration,ASC mRNA level was reduced by P.gingivalis.However,mRNA expression of NLRP3 was not changed.Meanwhile,mRNA expressions of NLRP3,ASC,Caspasel in inflammatory tissues were almost equal to those in normal gingival tissues,while mRNA level of IL-1β in inflammatory tissues was higher than that in normal tissues.In 24h and 48h,the protein levels of IL-1β in THP-1 cells were increased positively along with the increasing concentration of P.gingivalis,while the protein levels were lower than those in control groups,but the difference was found only in 24h.The protein level of NLRP3 showed no difference between treated and control groups.Conclusion:the apoptotic effects of P.gingivalis to THP-1 cells depend on the stimulating concentration and time;P.gingivalis could stimulate the secretion of IL-1β,and IL-1β levels were positively correlated with P.gingivalis stimulating concentration and time.P.gingivalis could just cause the expression changes of two targets of NLRP3 inflammasome,not NLRP3 itself.And further study is still needed.Objective:to observe clinical outcomes of combined and sequential treatment methods for patients with aggressive periodontitis,and to evaluate the therapeutical effect of basic periodontal therapy and its combination with GTR or orthodontic treatment to periodontal tissue regeneration.Methods:3 typical cases of aggressive periodontitis were chosen,which involved different treatments such as GTR,orthodontic treatments on the basis of basicperiodontal therapy.During the whole treatment processes,regular periodontal maintenance was necessary,clinical periodontal tissue conditions were examined and statuses of alveolar bone were evaluated before and after treatment with the application of X-rays or CBCT.Results:with the completion of comprehensive and sequential treatments of the above 3 cases with aggressive periodontitis,the periodontal statues were improved and there was no progression of new lesions.The satisfactory effects of function and aesthetics were achieved.Periodontal regeneration was promoted to some different extents.Conclusion:comprehensive and sequential treatments can promote periodontal tissue regeneration to a certain extent,and they can effectively treat aggressive periodontitis. |
