Objective:To observe the effect of Qishao granules on serum inter leukin-6(IL-6),interleukin-12(IL-12),thyroglobulin antibody(TGAb)and thyroid peroxidase antibody(TPOAb)in rats with experimental autoimmune thyroiditis(EAT),and to explorethe mechanism of Qishao granules against Hashimoto’s thyroiditi s.Materials and Methods:Sixty female SD rats of SPF grade were used,their age w as from 4 to 6 weeks and their weight was from 100 grams to120 grams.Adaptably fed for one week,15 rats were randomly selected as the normal control group,remaining for the mode ling group.Starting from the second week,the normal control group was fed purified water and the modeling group was fed iodine water whose concentration was 0.64g/L.Starting from the fourth week,the modeling group was injected with Tg and CFA in the feet subcutaneous twice in the week,each time ev ery rat was injected Tg of 100 ug for primary immune.Startin g from the fifth week to the eighth week,the modeling group was injected with Tg and IFA in the back subcutaneous once every week,each time every rat was injected Tg of 100 ug for strengthening the immune.After the EAT model was created in the eighth week,the modeling group was randomly divided int o three groups that were the model group,the tripterygium w ilfordii polyglycoside group and Qishao Granules group.Begi nning from the ninth week,the tripterygium wilfordii polygl ycoside group was given tripterygium polyglycoside tablets 6.25 mg per kilogram,Qishao Granules group was given 21.4g p erkilogram,the normal control group and the model group wer e given the same amount of saline solution.After lavage tre atment of four weeks,drawed blood from abdominal aorta of r ats and used enzyme-linked immunoassay(ELISA)to detect the levels of rat serum IL-6,IL-12,TGAb,TPOAb.Results:1.Compared with the normal control group,the levels of rats serum thyroid autoantibodies TPOAb and TGAb in the mod el group were significantly higher,there was a significant difference(p<0.01);compared with the model group,the leve ls of rats serum thyroid autoantibodies TPOAb and TGAb in Q ishao Granules group were significantly lower,there were a significant difference(p<0.01);compared with the triptery gium wilfordii polyglycoside group,the levels of rats serum thyroid autoantibodies TPOAb and TGAb in Qishao Granules gr oup did not change significantly,there was no significant d ifference(p>0.05).2.Compared with the normal control group,the levels of rats serum IL-6 and IL-12 of the model group were significa ntly higher,there were a significant difference(p<0.01);compared with the model group,the levels of rats serum IL-6 a nd IL-12 in Qishao Granules group were significantly lower,there were a significant difference(p<0.01);compared with the tripterygium wilfordii polyglycoside group,the levels o f rats serum IL-6 and IL-12 of Qishao Granules group did no t change significantly,there was no significant difference(p>0.05).Conclusion:1.Qishao Granules can significantly reduced the levels of serum thyroid autoantibodies TGAb and TPOAb in EAT rats.2.In reducing the levels of serum thyroid antibodies TG Ab and TPOAb in EAT rats,Qishao Granules compared with tri pterygium polyglycoside tablets,there was no significant d ifference.3.Qishao Granules can significantly reduced the levels of serum IL-6,IL-12 in EAT rats.4.In reducing the levels of serum IL-6 and IL-12 in EAT rats,Qishao Granules compared with tripterygium polyglycos ide tablets,there was no significant difference.5.The treatment of Qishao Granules and tripterygium pol yglycoside tablets was equivalent.. |
