Ulcerative Colitis(UC)is a chronic non-specific inflammatory bowel disease characterized by unexplained chronic inflammation and ulceration of colonic mucosa.Because of its etiology and pathogenesis is unknown,and the lack of effective treatment of drugs and methods,making the course of the disease more difficult.Compound bitter soup for our hospital treatment of UC clinical experience side,by the bitter ginseng,green Dai,Bletilla,to the four Chinese herbal composition,with bleeding to stop bleeding,swelling and convulsions,detoxification,dampness,insecticide,cooling blood to stop bleeding The efficacy of the drug for the treatment of UC more than a decade,to achieve good results.In this study,we will use the modern key technology of traditional Chinese medicine to study the extraction and isolation of effective parts and main active components of compound bitter soup,and use 2,4,6-trinitrobenzene sulfonate(TNBS),And to explore the effective part of the treatment of compound bitter soup by using immunohistochemistry and molecular biology.The mechanism of the main active ingredients was discussed,and the effect of compound Fufang Kushen decoction was studied.Pharmacological basis of the material and the selection of active lead compounds to provide a scientific basis.This paper mainly completes the following research work.Using the key technology of traditional Chinese medicine to extract the different chemical parts of the compound bitter soup,the UC rat model was used to screen the above-mentioned chemical site for the screening of UC activity,and the main active site was determined.The chemical components of the main active sites were separated and identified by means of modern chromatographic techniques such as silica gel column chromatography,gel chromatography and distribution chromatography.Using the cell pharmacology test,molecular biology method,the chemical composition of the treatment of UC activity screening,determine the main active ingredients.The effect of active ingredient on the expression of tight junction protein was detected by Western blot.The results showed that compared with the LPS group,the active ingredient group can increase the expression of tight junction protein,resulting in anti-UC effect.
