Bidens bipinnata L.is an annual herb and belongs to the Bidens genus(Asteraceae),which includes about 240 species with cosmopolitan distribution.Bidens bipinnata L.grows to a height of up to 1.5m with green leaves,yellow flowers,tiny black seeds and widely distributed in disturbed areas and along roadsides in tropical and subtropical climates.It was known worldwide as traditional medicine for the treatment of cold,malaria,diabetes mellitus,jaundice and so on.In recent years,researchers have performed a lot of research on the biological activities and chemical constituents of Bidens bipinnata L.,which showed that it has capacity to anti-malaria,anti-inflammatory,anti-tumor,anti-liver fibrosis,anti-oxidation and so on.And research showed that Bidens bipinnata L.contains polyacetylenes,acids,flavonoids and so on.It has large amounts of flavonoids,and most of them have 2-phenyresearl-chromone,and further research indicated many compounds have good biological activity.Objective: To isolate,purify and identify the chemical constituents of the total flavonoids of Bidens bipinnata L.and screen for their Bio activities.Methods: The constituents of the total flavonoids of Bidens bipinnata L.were isolated by reverse-phase silica gel column chromatography,following by Sephadex LH-20,MCI and TLC.The structures were identified by NMR.(1H-NMR and 13C-NMR).The inhibition for the proliferation of HSC-T6 cells was evaluated by MTT.The HSC-T6 cells and RAW264.7 cells were cultured and stimulated with the compounds with the concentrations ranging from 6.25 to 100μmol/L.The cell proliferation ability was detected by MTT method,and the concentration of tumor necrosis factor α in the supernatant of RAW264.7 cells was measured via ELISA.Results: Twelve components were isolated and identified as ethyl caffeate(1),iso okanin(2),Threo-dihydroxydehydrodiconiferyl alcohol(3),octacosane(4),hesperidin(5),naringenin(6),apigenin(7),keampferol-3-O-α-l-rhamnoside(8),luteolin(9),malto se(10),gallic acid(11),hyperin(12).Compounds 1 and 3 were isolated from thisg enus for the first time.Components 9 and 11 could inhibit the proliferation of H SC-T6 cells and decrease the TNF-α concentration in the supernatant of RAW264.7 cells.Conclusion: Twelve compounds were isolated from the Bidens bipinnata L.,among which compound 1 and 3 were first isolated.Compound 9 and 11 have potent bio activities in vitro.
