Obejective: Deregulated SOCS3 is a characteristic feature of acute leukemia.Our study wanted to learn whether SOCS3 expression is altered in pediatric patients with acute lymphoblastic leukemia(ALL),and to investigate the relationship between SOCS3 expression level and patients clinical characters,and finally offered a new method for the evaluation of ALL risk and treatment for ALL.Methods We therefore quantified SOCS3 expression in pretreatment and complete remission bone marrow samples from pediatric patients enrolled and correlated SOCS3 expression with patient demographics,laboratory characteristics and clinical outcome.Besides,immunofluorescence histochemistry and laser confocal microscopy were applied to analyze the expression level of SOCS3 in ALL patients bone marrow samples.Results The results showed SOCS3 decreased in pretreatment and increased to normal level in complete remission.The decreased SOCS3 finally attributes to the sustained phosphorylation and expression of STAT3.Relative high SOCS3 expression in pretreatment indicated higher risk.Conclusion SOCS3 is associated with ALL risk and could serve as a prognostic or predictive marker for ALL stage and risk regulation,and SOCS3 could be used as an indicator of ALL states.It remains untested whether high SOCS3 expression identifies a subset of leukemia with deregulated cytokine signaling that could be amenable to therapeutic intervention.
