Ganoderma lucidum spore powder is a spore of Ganoderma lucidum fungus.The Ganoderma lucidum as a medicinal fungus has long history of its medicinal efficacy.Nearly 40 years,Ganoderma lucidum is widely used for prevention and treatment in chronic bronchitis,hyperlipidemia,hypertension,neurasthenia,hepatitis and other diseases prevention and treatment[1].Ganoderma lucidum spore powder is Ganoderma in its fruiting body after the development of mature,constantly from the bacteria tube shot out of spores collected.Its medicinal value is much higher than the Ganoderma lucidum fruiting body and mycelium.However,Ganoderma lucidum spores have double walls,the general chemical and physical methods is difficult to break the double layer of Ganoderma lucidum spores,thus limiting its chemical composition,bio-active ingredients in-depth study.With the further development of technology,people use a variety of effective ways to break down the Ganoderma lucidum in order to better study it.In this paper,Ganoderma lucidum spore powder as the research object,mainly on its chemical composition research.First,the Ganoderma lucidum spore powder extraction with methanol solution 3 times,combined extract,concentrated under reduced pressure,obsolete Ganoderma lucidum spore powder extract,weighing,after the appropriate amount of distilled water dissolved into a suspension,successively partitioned and extracted with petroleum ether,ethyl acetate and n-butanol,concentrated layers of extract.Comprehensive use of a variety of chromatography separation methods,such as silica gel column chromatography,reverse phase silica gel column chromatography,high and medium pressure preparation chromatography,mainly on the petroleum ether,ethyl acetate,the two extraction layer for further separation and purification.The final separation of 10 monomer compounds.Wherein four compounds were isolated from the petroleum ether layer extract and six compounds were isolated from the ethyl acetate layer extract.According to the physical and chemical properties and,NMR techniques and with the standard controls with the final analysis of 10 kinds of compounds in the structure.They are:Oleic acid(1),Ergosterol-7,22-diene-3β-ol(2),B-sitosterol(3),Cerevisterol(4),β-daucosterol(5),α-Tocophero(6),(2R,3S)-2.3-dihydro-2-(4’-hydroxy-3 ’-methoxy-phenyl)-3-hydroxylmethyl-7-methox y-5-benzofuran-propapol-4’-O-β-glucopyranoside(7),Mannitol(8),β-D-fructofturanosy-α-D-Allopyranoside(9),Sucrose(10)。In addition,in order to more fully understand the pharmacological effects of Ganoderma lucidum spore powder.The results showed that the protective effect of Ganoderma lucidum spore powder methanol extract on acute liver injury in mice was studied,It was found that Ganoderma lucidum spore powder had a good pharmacological effect on glutamate aminotransferase(ALT),malondialdehyde(MDA)and superoxide dismutase(SOD),and through the mouse liver tissue HE staining found that CC14 acute liver injury have a protective effect.Finally,the petroleum ether layer extraction of Ganoderma lucidum spore powder was analyzed by GC-MS for volatile components.The results of a total of more than 85%of the total of 75 kinds of ingredients.Which are mostly aliphatic compounds,fatty acids and fatty alcohols,compared with the results of GC-MS of Ganoderma lucidum spores in literature[2]-[4]。The GC-MS results of this paper have more volatile components in Ganoderma lucidum spore powder.More comprehensive.
