Slow transit constipation(STC)is a clinically common chronic intractable constipation,accounting for 25%of the incidence of constipation.Its clinical manifestations is stubborn,and the effect of comprehensive conservative treatment is unsatisfactory.Due to clinical abuse of laxatives,leading to progressive increase in constipation,and ultimately lead to drug failure can only be carried out surgical treatment.The pathogenic mechanism of slow transit constipation is still not clear,which brought great difficulties to its clinical prevention and treatment.In this study,molecular biology methods combined with high-throughput sequencing were used to analyze the microbial community structure of stool samples in slow transit constipation and in the process drinking mixed fermented yoghourt of probiotics.The physiological indexes of STC mice were analyzed,to study the effect of mixed fermented yoghourt on STC,and in order to provide a theoretical basis for clinical diagnosis and treatment.Experimental results showed that the three bacteria of yoghurt can significantly alleviate the symptoms of STC.Through the detection of physiological indicators of mice,in the three bacteria of yoghurt,the small intestine propulsion,the first row of black time is better than other groups,and it can significantly reverse the symptoms of constipation in mice.The detection of short-chain fatty acids(SCFAs)in mouse stool samples shows that the three bacteria of yoghurt has a significant effect on reducing the energy supply.From the number of OTUs,the ACE index and the Chao]index,the bacterial diversity of the stool samples in mice and slow transit constipation patients was significantly reduced with the days increase of feeding yoghurt,while after fed with constipation drug,the number of OTUs in the control group and two bacteria of yoghurt group was significantly higher,but there was no significant difference between the two groups.While the three bacteria of yoghurt group and constipation group were descreased.The bacterial diversity in stool samples of patients with slow transit constipation is significantly reduced,indicating that constipation leads to an increase in harmful microbes in the intestine,which may result in inhibition of some other microbial growth and even death.After feeding mixed fermented yoghourt of probiotics,the beneficial microorganisms are colonized in the intestine,inhibit the growth of harmful microorganisms,so that the diversity of bacteria in the stool samples shows a decreasing trend.In the mouse experiment,compared the bacterial community structure of all the mice in different groups showed that the major bacteria in the four groups of the stool samples were Bacteroides,Firmicutes and Proteobacteria,in the phylum level.The amount of Firmicutes and Proteobacteria in the two bacteria of yoghurt group was significantly lower than other groups,while the proportion of Bacteroides was higher.With the days increase of drinking yoghurt,the microbial community in the stool samples of each group showed obvious changes.The microbial community structure analysis by differet treatment time showed that Candidatus family ratAN060301C and Rikenellaceae increased with the days increase of feeding yoghurt,while Lachnospiraceae,Desulfovibrionaceae and Carnobacteriaceae were decreased,the reason why its increase or decrease in the stool sample is due to its change in the intestine.UPGMA(unweighted pair-group method with arithmetic means)and PCA(principal components analysis)analysis showed that,regardless of the method,the results of the bacterial communities clustering are roughly the same,the distribution of bacterial community in the stool samples of the pre-fed probiotics was not uniform and no significantly correlation,while the bacterial community structure was similar after feeding yoghurt,and the three strains were more similar to the two strains.The results showed that the bacterial community structure was similar in the stool samples after feeding the probiotics,and the three bacteria of yoghurt group were more similar to the two bacteria of yoghurt group.Compared the bacterial community structure of the fecal samples before and after drinking mixed fermented yoghourt of probiotics,it can be seen that the bacterial diversity in the fecal samples before the yoghurt was higher than that after,at the level of phylum.Nearly half of the sequences belong to Bacteroidetes and Firmicutes,while Proteobacteria and Actinobacteria were less.At the level of genus,the bacterial diversity in the stool samples before and after drinking mixed fermented yoghourt of probiotics were no significant difference,and were Bacteroides and Faecalibacterium.Through the analysis and comparison of the major bacteria in the stool samples before and after yoghourt at the level of genus,the majority of patients before and after drinking mixed fermented yoghourt was Bacteroides,most of the fecal,most of them was Faecalibacterium.By comparing the intestinal microbial community structure and succession discipline of slow transit constipation patients before and after drinking mixed fermented yoghourt of probiotics,combined with slow transit constipation mice model test,looking for the key micro-organisms which were in constipation and the process of relieve constipation by mixed fermented yoghourt,and for the clinical treatment of constipation to provide a powerful new treatment.
