| Objective:A rat model of lipopolysaccharide induced acute lung injury is us-ed to study the effect of polysaccharide from armillaria mellea on acute lung inju-ry in rats and its related mechanism.Methods:35 healthy male SD rats(clean)were randomly allocate into five groups:norma groupl,model group,positive control group,low and high polysaccharide from armillaria mellea(200mg/Kg and 400mg/Kg)dose group with 7 rats each.Respectively,medication group gavage low and high doses of polysaccharide from arimillaria mellea for 10 days,the other groups were given the same dose of saline.After 10 days,two hours before the injection of LPS to,positive control group receive the dexamethasone(5mg/Kg)intraperitoneal injection of medication.After two hours later,all the rats with intraperitoneal injection of lipopolysaccharide(20mg/Kg)induced acute lung injury except the normal group.Then the rats in nomal groups were control intraperitoneally injected same volume PBS.After lipopolysaccharide injection of 6h,All rats were executed by drawing-out all the blood in their hearts with 20%urethane-anesthetized.Detection index:(1)Lung organization pathology learn observation;(2)used ELISA method detection plasma in the endothelial pigment-1(ET-1),and thrombosis regulation protein(TM),and interleukin-6(IL-6),and Tumor necrosis factorα(TNF-α)of content.(3)with protein immune imprinted test(Western Blot)detection the group p-PI3K,and p-Akt,and NF-κ b,and p-NF-κb,and Ik-B expression,determination the article with grayscale value.Results:1.Lung biopsy shows,after lipopolysaccharide injury,alveolar damage,and alveolar septal thickening,vascular cavityhemorrhage and micro thrombosisn;positive control and low dose groups and high-dose groups compared with the model groups,alveolar structure is more complete,alveolar septal thinning,vascular bleeding and micro-thrombosis cut back.2.The results of ELISA showed that compared with the normal group,the levels of TNF-a,IL-6,TM and ET-1 in the model group were significantly higher(P<0.05);Compared with the model group,the levels of TNF-a,IL-6,TM and ET-1 in high dose group was significantly decreased(P<0.05),the levels of TM and ET-1 in low dose group was significantly decreased(P<0.05).3.Western blotting showed thatcompared with the normal group,the expression of p-PI3K,p-Akt,NF-κ B and p-NF-κ B in the model group were significantly higher(P<0.05),and the expression of Ik-B was significantly decreased(P<0.05);Compared with the model group,the expression of p-PI3K,p-Akt,NF-κ B and p-NF-κ B in the polysaccharide from armillaria mellea group were significantly decreased(P<0.05),and the expression of Ik-B was significantly higher(P<0.05).Conclusion:1.Polysaccharide from arimilllaria mellea has a protective effect on acute lung injury in rats induced by LPS.2.PI3K/Akt/NF-κ B signaling pathway may participates in the protection of polysaccharide from armillaria mellea to acute lung injury induced by lipopolysaccharide(LPS)in rats. |
