Objective:Study on the chemical constituents from Tetrastigma planicaul(Hook.)Gagnep.This work can give a scientific basis for ex-posing the antitumor activity extracts from the plant.Methods:T.planicaul was extracted five times by 80%ethanol,and the extract was separated into five parts according to the polarity, including petroleum ether part, ethyl acetate part,n-butanol part and wa-ter part. The constituents were isolated by silicagel, macroporous resin adsorption, polyamide,Sephadex LH-20 chromatography, recrystallization,et al.The structure were identified by spectral analysis(MS,1H-NMR,13C-NMR et al)and comparing with the published data. The active ingr-edient protocatechuic acid of T.planicaul was qualitatively identifiedby TLC,and quantitative dermined by HPLC.Result:Twenty-four compounds were isolated and identified as coelarthenol(1),ethyl 3β-hydroxy-5-cholen-26-oate(2),taraxerone(3),β-sitosterol(4),daucosterol(5),protocatechuic acid(6),dennettine(7),ethyl gallate(8),tetradecanoic acid(9),1-dodecanol(10),glyceryl pal--mitate(11),3-β(stearyolxy)olean-12-ene(12),palmitic acid(13),salicy-lic acid(14),β-sitosteryl oleate(15),vanillic acid(16),stigmasterol(17),12-methyltridecan-1-ol(18),(24s)-3β-hydroxy-5α-stigmastan-6-one(19),isoscopletin(20),estigmast-4-en-6β-ol-3-one(21),stigmast-4,2,2-dien-3-one(22),D-glucose(23),D-mannitol(24).Content of protocat--echuic acid of T.planule had a good linearity in the range of 0.02920.584μg(r=0.9998).The result of methodology validation meets the re-quirement.Conclusion:Compound 13,7,9,10,12,15,1822 are iso-lated from Tetrastigma BI.for the first time.13,7,9,10,12,15,1824 are obtained from this plant for the first time. The method can be used for the estimation of quality of T.planule.
