The Effects Of Wulongdan On NF-κB Singnaling Pathway In Rheumatoid Arthritis

Objective:To statistically analyze the influence of Wulongdan on cell factors(NF-κBp65)and inflammatory factors(IL-1,TNF-α,IL-6,IL-8)in serum of RA sufferers and further explore its action mechanism on rheumatoid arthritis(RA)of type of obstruction of phlegm and blood stasis.Methods:Divided the 60 sufferers meeting inclusion standard into treatment group and control group according to the principle of random control,with 30 sufferers for treatment group,who orally took Wulongdan three times a day,20 pills each time,which method was combined with leflunomide tablets,20mg each time and once every day.30 sufferers belonged to control group,who took leflunomide tablets,20mg each time and once every day.The treatment cycles were all 12 weeks,during which period,the two groups were observed for clinical symptoms.Before treatment,statistical analysis shows that the two groups of sufferers were comparable in gender,age,and course of disease,etc.Observed and recorded the changes of TCM symptom integral of the two groups before and after administration to evaluate the curative effect for TCM symptoms.Observed and recorded each related lab index of the two groups before and after administration(used Elisa method for detection):the change in value of NF-κBp65,IL-1,TNF-α,IL-6 and IL-8 in peripheral serum.Conducted safety monitoring for their liver and kidney functions,abnormity of blood and urine routine examination(detected the condition for the 0th,4th,8th and 12th week)Results:1.In terms of curative effect for TCM symptoms,the treatment group outperformed the control group(P<0.05).2.Comparison of score of each TCM symptom:in terms of improving arthralgia,turgidity,tenderness,sluggishness of joint movement,perspiration,aversion to wind and cold,soreness and weakness of waist and knees,morning stiffness time,etc.,the treatment group outperformed the control group(P<0.05).In total integral of TCM symptoms,the treatment group outperformed the control group(P<0.05).3.In lab indices:the two groups’ NF-κBp65,IL-1,TNF-α,IL-6 and IL-8 all improved markedly than those before treatment.After treatment,the control group outperformed the control group,with difference having statistical significance(P<0.05).Conclusion:After statistical analysis,it was concluded that treatment of rheumatoid arthritis(RA)of type of obstruction of phlegm and blood stasis using Wulongdan delivers definite clinic curative effect,and can significantly reduce the level of NF-κBp65,IL-1,TNF-a,IL-6 and IL-8 in serum.The action mechanism is down-regulating the content of related cell factors by regulating NF-κB signaling path to remit arthritis symptoms of RA sufferers.
