At present,many studies have shown that the underlying cause of inflammatory bowel disease(IBD)may be associated with excessive inflammation caused by intestinal tract disorders that impair intestinal tolerance.The role of intestinal flora is the direction of enteritis target research,cinnamon essential oil is cinnamon volatile oil extract,it has the role of inhibition of pathogen growth,and inflammatory bowel disease and human intestinal microbial imbalance.And cinnamon essential oil on intestinal flora imbalance caused by the treatment of inflammatory bowel disease is not clear.In this study,the effect of subcritical water extraction on the efficacy of cinnamon essential oil was studied in this experiment.By controlling the model of enteritis mice,the body weight of colonitis mice,colon length and pathology,myeloperoxidase(MPO)activity,hemoglobin content and immune organ index detection,indicating the role of cinnamon essential oil.Furthermore,the DNA V3-V4 region of each group was analyzed based on the high throughput sequencing technique of Illumina MiSeq sequencing platform,and the effects of cinnamon essential oil on the bacteria of enteritis mice were analyzed.The results of the study will reveal the mechanism of cinnamon essential oil to reduce enteritis from the perspective of intestinal flora,and provide a new therapeutic target for enteritis treatment.1.Cinnamon essential oil to alleviate the ability of inflammatory bowel diseaseIn this study,the effect of cinnamon essential oil on inflammatory bowel disease was studied from the perspective of changes of mouse signs.The mice were randomly divided into normal group,antibiotic group(10 mg/kg/day of ceftriaxone sodium),enteritis group(3%DSS)and cinnamon oil group(high dose group 15 g/kg,low Dose group 10 g/kg,sigma group 10 g/kg),each group of 6.The changes of body weight and signs were observed daily and the DAI score was performed.At the end of the model,the length of the colon was measured and the histopathological analysis was performed.The activity of myeloperoxidase(MPO)was measured.The immune organ index was detected by thymus and spleen.The hemoglobin content was measured by blood sampling.The mice of colitis model were given cinnamon essential oil and antibiotics respectively by intragastric administration,and the pharmacodynamic evaluation was carried out.The results showed that cinnamon essential oil alleviated the symptoms of DSS in mice,colon injury,myeloperoxidase(MPO)activity,spleen index increased,thymus index and colon index decreased hemoglobin content and other symptoms.Compared with enteritis group,the body weight of thecinnamon oil group increased,the DAI index decreased,the colon length increased,the pathological sections of colon tissue were infiltrated in inflammatory cells,and the structure of crypt was improved.The DAI index was reduced by a factor of 3,the length of the colon increased by about 16%,the activity of myeloperoxidase(MPO)was doubled,and the effect of low-dose and sigma groups was not significant.(45.33 vs 63.87,P<0.05),and the MPO index was significantly decreased(1.33 vs 0.57,P<0.05).The effect of antibiotics on the treatment of inflammatory bowel disease was significantly higher than that of the control group(45.33 vs 63.87,P<0.05).2.Based on high-throughput analysis of cinnamon essential oil on enteritis mice intestinal floraIn this experiment,we also try to discuss the treatment mechanism of cinnamon essential oil from inflammatory bowel disease by changing the direction of intestinal flora.Based on the Illumina MiSeq sequencing platform,the V3-V4 region of mouse fecal DNA was subjected to high-throughput sequencing to analyze the changes of intestinal flora diversity.At the gate level,Firmicutes and Bacteroidetes were the dominant dominant bacteria in the normal group,accounting for 48.77%and 48.03%of the total bacteria,followed by Proteobacteria.Compared with the normal group,the dominant bacteria status of Firmicutes and Bacteroidetes in DSS-induced feces was not changed,but Firmicutes decreased,the number of Bacteroidetes increased and the number of Proteobacteria changed significantly.The total number was higher than that of the normal group.The total number of bacteria in the intestine was 10.76%.After treatment with cinnamon oil,the intestinal microflora structure of the enteritis group was significantly different,the number of Bacteroidetes and the Proteobacteria were decreased(10.76%vs 0.56%,P<0.05),the number of Firmicutes increased,and the effect of the high-dose group was significant cut back.At the level of the department,the number of Bacteroides species was significantly close to the normal level after treatment with cinnamon oil,and the content of S24-7 was significantly increased.The treatment effect of high dose group and low dose group showed obvious difference(44.06%vs 27.7%,P<0.05).In the form of Heatmap,the specific flora of response to cinnamon essential oil was discussed,and the heatmap of genera were analyzed.The results showed that the enteritis group returned to normal after the treatment of enteritis mice with cinnamon essential oil,and the effect of the high dose group of cinnamon oil was significant,the fecal bacteria(1.07%vs 0.14%,P<0.05),Bacteroides(33%vs 9.01%,P<0.05)and lactic acid bacteria(4.24%vs 0.32%,P<0.05)decreased significantly,and after antibiotic treatment,Intestinal flora disorders.The results showed that the Bacteroidales_S24-7_group in the Clostridiales of the enteritis group was significantly decreased(32%vs 21%,P<0.05)and the Lachnospiraceae_NK4A136_group in the bacillus(29%vs 13%,P<0.05)was significantly decreasedby LEfSe.After treatment with cinnamon essential oil,the strains returned to normal levels.In summary,this experiment confirmed that inflammatory bowel disease and intestinal flora is closely related to,and cinnamon essential oil can alleviate intestinal microbial disorders,improve intestinal microbial abundance,by increasing the intestinal structure of mice diversity,Intestinal stability of the environment,play a preventive and improve the effect of inflammatory bowel disease,for the treatment of inflammatory bowel disease provides a new way of thinking.
