| Objective:1.TRPM8 ion channels can provide guidance to estimate the malignant degree and prognosis of pancreatic cancer as clinical biomarkers.2.TRPM8 ion channels as potential therapeutic targets and may silence its genes to control the proliferation and spread of the pancreatic cancer cells in order that we can discover the new targets and new way for personalized therapies in pancreatic cancer.Methods:1.77 cases of pancreatic cancer were purchased from Beijing dragon Midas science and Technology Development Co.,Ltd.and conducting experiment with immunohistochemistry technology and analyzing the results.2.Buying normal pancreas cell lines HPCY-5,pancreatic cancer cell lines Bx PC-3 and PANC-1,and detecting the TRPM8 ion channels m RNA expression in that three kinds of cell lines with QPCR.3.To detect the channel protein expression of HPCY-5,Bx PC-3 and PANC-1with Western Blot technology.4.Using patch clamp technique to detect the electric current change of cell lines PANC-1 before and after transfection with si RNA.5.Using CCK8 kits to research the proliferative capacity of pancreatic cell lines Bx PC-3 and PANC-1 before and after transfection with si RNA.Results:Both Immunohistochemical results and Western blot results suggest that TRPM8 ion channels protein is highly expressed in pancreatic cancer cells,andImmunhistochemical results shows that the over-expressed of TRPM8 channels ptotein is correlated with the degree of tumor differentiation(middle/low differentiation versus high differentiation P<0.05,the difference was statistically significant).QPCR detect that the TRPM8 ion channels m RNA level of pancreatic cancer cell lines Bx PC-3,PANC-1 is higher than that of normal cell lines HPCY-5,which is consistent with the results of immunohistochemistry and Western Blot,and P<0.05,the difference was statistically significant.Patch clamp technique shows that menthol activated TRPM8 ion channels current increased significantly.CCK8 assay indicate that the proliferation ability of TRPM8 gene knockout cancer cell lines is significantly weakened,which showes that the expression of TRPM8 channels gene has an effect on the increase of cancer cells. |
