| Objective: We proposed to find the invasive related important proteins with nonfunctional pituitary adenomas.These proteins to the mechanism of nonfunctional pituitary adenoma invading growth provide theoretical basis for the comprehensive and systematic study,This study using the method of proteomics may provide a new breakthrough for the system research of nonfunctional pituitary adenomas pathogenesis and its molecular target therapy.Methods: Collected 18 cases nonfunctional pituitary adenomas patient’s tumor specimens which in Xinjiang uygur autonomous region people’s hospital neurosurgery during October 2015 to October 2016(9 cases of invasive growth,9 cases of noninvasive growth).We are looking for the nonfunctional pituitary adenomas different proteins expression by use two-dimensional gel electrophoresis and mass-spectrometric technique.We screen the different express proteins from invasive nonfunctional pituitary adenomas and noninvasive nonfunctional pituitary adenomas by use two-dimensional gel electrophoresis(2-DE)and Matrix assisted laser desorption ionization time of flight mass spectrometry(MALDI-TOF-MS).To move forward a single step on those proteins which may be applied in the clinical diagnosis.Results:(1)By the two-dimensional electrophoresis results(Through PDQuest software show the differentially expressed proteins spots),we constructed of nonfunctional pituitary adenomas of 2-DE map.(2)Through the research we found four spots of different proteins,on this basis,the protein of p14 ARF in the invasive nonfunctional pituitary adenomas tumor tissues down-regulates expression,the proteins of EGFR、HMGA2、PIK3CA in the invasive nonfunctional pituitary adenomas tumor tissues up-regulate expression.Conclusion: The 4 kind different expression proteins have possible relationship with the invasiveness of nonfunctional pituitary adenomas. |
