| Objective:To preliminary discuss the effect of moxibustion on AMPK and mTOR in rat myocardial injury of an exhaustive exercise.Methods:24SD male rats were randomly divided into blank control guoups(n=8),exhaustive exercise guoups(n=8),and the preventive moxibustion groups(n=8).Exhaustive exercise guoups without any treatment in the fist 10 days that had been sampled organization in the eleventh day after exhaustive exercise.Preventive moxibustion group were applied to "Zusanli"(ST 34)and"Guanyuan"(CV 4),once daily for 10 days,sample organization on the 11th after exhaustive swimming.The two groups of rats were recorded to achieve exhaustive use of time.HE staining was used to observe the pathological changes of myocardial tissue;the serum LDH was detected by colorimetric method;the content of CK-MB in serum was detected by immune suppression method;the content of cTnT in serum was detected by Elisa;the content of AMPK/mTOR in myocardial tissue was detected by Western blot.Results:(1)Time:Compared with the exhaustive exercise group,the exhaustive exercise time of the preventive moxibustion group was increased(P<0.05).(2)HE staining:HE staining was found to have a clear structure of the myocardium in the blank control group and preventive moxibustion group,and the inflammatory cell infiltration was reduced in preventive moxibustion group.There was a little inflammatory cell infiltration in the exhaustive exercise group.(3)LDH,CK-MB,cTnT content in serum of every groups:Compared with the blank control group,the serum LDH,CK-MB,cTnT content of exhaustive exercise group were increased(P<0.05);Compared with the Exhaustive exercise group,the serum LDH,CK-MB,cTnT content of preventive moxibustion group were increased(P<0.05);(4)MDA and SOD content in serum of every groups:Compared with the blank control group,the serum MDA content was increased and SOD content was decreased(P<0.05);Compared with the Exhaustive exercise group,the serum MDA content was decreased and SOD content was increased(P<0.05);(5)The AMPK and mTOR content in myocardial tissue of every groups:Compared with the blank control group,myocardial tissue AMPK content was increased(P<0.05),and the myocardial tissue mTOR content was decreased.Compared with the Exhaustive exercise group,the AMPK content in myocardial tissue was increased,and the mTOR content was decreased(P<0.05)and AMPK and mTOR were negatively correlated(r=-0.764 P=0.01<0.05 bilateral).Conclusion:Effect of rats with myocardial injury in preventive moxibustion anti-exhaustive exercise maybe based on AMPK/mTOR Signaling Pathway. |
