| Objective:1.To study the content of polysaccharides and alcohol extracts of Dendrobium huoshanense and its main products in different growth years,and to explore the difference between the Dendrobium huoshanense and Dendrobium candidum and the best recovery period.2.To study the difference of polysaccharide and polyphenol content in Dendrobium candidum under different light conditions,the optimum light conditions of Dendrobium candidum were studied.3.The GC-MS study of polysaccharides from Dendrobium huoshanense,Dendrobium candidum and Dendrobium candidum was studied by different methods.4.Study on the first-order spatial structure of polysaccharides from Dendrobium huoshanense,and provide the basic data for the further study of structure-activity relationship.Methods:1.The polysaccharides of Dendrobium huoshanense,Dendrobium and its main mixed products were studied by 95% alcohol extraction and then alcoholic acid precipitation,anthrone-acid acid coloring and drying weighing.And the content of alcohol extracted,to determine the difference between the two and the best harvest period,from the chemical point of view to reveal the best effect of Dendrobium huoshanense the reasons for the better.2.The differences of polysaccharides and polyphenols in Dendrobium huoshanense were compared by two kinds of different illumination of LED,red and blue light in two groups of the same kind of tissue.3.Dendrobium huoshanense and several other’s Dendrobium of polysacchar idesrefined by hydrochloric acid methanol After acid hydrolysis,the monosaccharide composition of several polysaccharides of Dendrobium were determined by TMS and GC-MS methods,and the difference of monosaccharide composited ofpolysaccharides was determined.4.The purified polysaccharides of Dendrobium huoshanense were decolored by polyamide column,homogeneous polysaccharides isolated from DEAE Bestarose FF and Sephacryl G-200 were analyzed by high performance gel column,GC-MS,FTIR and NMR,Glycosidic linkages and functional group properties to determine its first-level spatial structure.Results:1.The contents of polysaccharides and alcohols in Dendrobium huoshanense were higher than those in Tissue culture Dendrobium huoshanense,and the contents of polysaccharides and alcohol extractiones in Dendrobium huoshanens,Dendrobium huoshanens and Dendrobium henanense were differented in different growth years.,2-year-old group of Dendrobium huoshanense,wild Dendrobium huoshanense and Dendrobium henanense polysaccharide and alcohol content of the highest.2.Under the experimental conditions,the total content of polysaccharides and polyphenols in Dendrobium huoshanense treated by LED light was significantly better than that of red and blue light treated Dendrobium huoshanenes,and the two groups were treated with different light treatments The contents of polysaccharides and polyphenols in Dendrobium huoshanense were significantly higher than those in roots and leaves.3.The content of monosaccharide in Dendrobium huoshanense and other Dendrobium officinale was significantly different.The contents of mannose and glucose in wild Dendrobium were 62.0 ± 1.2% and 27.0 ± 11.1% respectively by GC-MS,,About 2 to 3 times the mannose and glucose content of Dendrobium huoshanense and Dendrobium officinale,and the content of galactose in Dendrobium officinale was 25.8 ± 11.3% much higher than that of Dendrobium officinale polysaccharide content(2.2 ± 10.9%).There were also significant differences in the content of monosaccharides in the other polysaccharides of Dendrobium officinale.4.Polysaccharide was isolated and purified to obtain a acidic homogeneous polysaccharide TC-DHPA4 with a molecular weight of 8.0 × 105 Da.The contents of rhamnose,arabinose,mannose,glucose,galactose and glucuronic acid were 1.28: 1:1.67: 4.47 : 10.43: 1.42 molar ratio.(1 → 4)-linked glucose,(1 → 4)-linked glucuronic acid,(1→4,6)-linked glucose,(1→6)-linked galactose,(1→3,6)-Linked galactose.Conclusion:The total contents of polysaccharides and alcohol extracts in Tissue culture Dendrobium and Dendrobium huoshanense were higher than those in Tissue culture Dendrobium,and the content of polysaccharides in Tissue culture Dendrobium and Dendrobium huoshanense was similar to that of other Dendrobiumes,In addition,there may be differences between the content of monosaccharides in the growth environment factors,tissue culture and wild Dendrobium officinale polysaccharides.The structural identification of the homogeneous polysaccharides of Dendrobium huoshanense was obtained,which provided the theoretical basis for the study of the structure-activity relationship of Dendrobium huoshanense. |
