Phellinus igniarius,a fungus belonging to the family Hymenochaetaceae,known as “Forest Gold”,which is one of the most important chinese traditional medicinal Phellinus fungi,preferably hosts on Morus alba L.,Populus L.,Quercus Linn,and other hardwood trees.Mainly distributed in Northeast China,North China,northwest and Sichuan,Yunnan and other places,in Japan,South Korea,North Korea and other East Asian countries are also distributed.Phellinus fungus is recognized internationally as the highest efficient fungus in the field of biological cancer therapy,has a wide range of medicinal value.As a kind of traditional Chinese medicine,it has been mainly used for treatment of cancer,liver diseases,and stomach ailmentsa.In recent years,the domestic and foreign researches indicate that Phellinus igniarius contain a variety of active ingredients.It has been reported that many compounds have been isolated from Phellinus igniarius,including terpenoids,steroidal,flavonoids,pyranones,phenoloic compounds,coumarins,polysaccharides and other species.Study on the pharmacological effects mainly display in: anti-tumor,immunoregulation,antiperoxidation,Hepatoprotective and antilivercirrhosis,and so on.In order to search the material of Phellinus fungi's antitumor and investigate precious Phellinus fungus resource in China,we studied the chemical constituents of natural Phellinus igniarius and the artificial culture of Phellinus fungus.In this paper,25 compounds were isolated from 95% ethanolic extract of the fruit body of natural Phellinus igniarius and the artificial culture of Phellinus fungus through a variety of methods and spectral analyses,such as HPD100 macroporous resin column,ODS,MCI,C8,silica gel thin-layer chromatography preparation,silica gel column chromatography,Sephadex LH-20 and NMR,mass spectrometry.Among them,12 compounds were isolated from natural Phellinus igniarius fungus as Uracil(1),ethyl-?-D-glucopyranoside(2),D-(+)-Trehalose(3),Furan(4),Uridine(5),Propanetriol(6),Cerebroside B(7),alismol(8),(22E,24R)-ergosta-7,22-diene-3?,5?,6?-triol(9),dimethyl phthalate(10),sakuranetin(11),(22E,24R)-ergosta-7,9(11),22-triene-3?,5?,6?-triol(12).From the artificial culture of Phellinus fungus,13 compounds were identified,including stigmasta-7,22-diene-3?,5?,6?-triol(13),paracetamol(14),4-(3,4-dihydroxyphenyl)-3-butene-2-one(15),protocatechualdehyde(16),3?-Hydroxyfriedel-2-one(17),3-Hydroxyfriedel-3-en-2-one(18),ergosta-4,6,8(14),22-tetraen-3-one(19),Ergosterol peroxide(20),Uracil(1),Uridine(5),Inotilone(21),inoscavinA(22),PhellibauminE(23).Compound(2),(3),(4),(8),(10)and(14)were isolated from Phellinus Quel.for the first time.And compound(9),(12)were isolated from Phellinus igniarius for the first time.The in vitro antitumor activity of compounds 15-23,1,5 was screened by CellTiter-GLo Reagent,on 41 human tumor cell strains and 2 hamster normal cell strains via high-throughput screening.Compounds 15,18,19,and 20 exhibit significant antitumor activity.In addition,all compounds had no toxic effect on normal cells.
