| Objective Find out miRNAs specific to Fas L by bioinformatics prediction.And to investigate its effect on the expression of Fas ligand(Fas L)in tongue squamous cell carcinoma,and finally to verify its effect on immune escape of tongue squamous cell carcinoma,So as to understand the tumor development and regulation mechanism.Methods The synthetic miR-590 mimics or miR-590 inhibitor was transfected into SCC-3 cells.The m RNA and protein expression of Fas L m RNA and protein in SCC-3 were detected by reverse transcription-polymerase chain reaction(RT-PCR)and Western blot;The transfected SCC-3 cells were treated with DDP(5ug / ml),The transfected SCC-3 cells were treated with DDP(5 ug / ml)and the cell growth curves were drawn at different time points by MTT assay.And the effect of miR-590 was verified from the negative side by the inhibitor of miR-590 against Fas L.Results The miR-590 mimics were transfected into SCC-3 cells.The total RNA was extracted 48 hours after transfection.The expression of Fas L m RNA and protein in SCC-3 cells were detected in the test group and control group.Mi R-590 was found to decrease the expression of Fas L on SCC-3 cells at m RNA and protein levels.However,the expression of Fas L m RNA and protein in SCC-3 cells transfected with miR-590 inhibitor was significantly higher than that in control cells.Add DDP by time gradient after SCC-3 cells were transfected with miR-590,the cell survival rate of experimental group is lower than the control group.Transfection of miR-590 inhibitor showed that the cell viability of the experimental group was higher than that of the control group.Conclusion Mi R-590 can effectively inhibit the expression of Fas L in SCC-3 cells and decrease the resistance of SCC-3 to DDP,which may reduce the apoptosis-inducing and killing effect of immunocompetent cells,and eventually block the way of the cancer cells to escape from the killing effect of the immune system,providing a new target for the treatment of tongue squamous cell carcinoma. |
