ObjectivesThe aim of this study was to detect the expression of platelet-derived growth factor-AA and phosphorylated extracellular signal-regulated kinase 1/2 in endometrial tissue of patients with intrauterine adhesions by immunohistochemistry,to investigate the possible mechanism of PDGF-AA and p-ERK1/2 in the pathogenesis of intrauterine adhesions(IUA),and to improve the clinical mechanism of PDGF-AA and p-ERK1/2 in the pathogenesis of intrauterine adhesions patients and the prevention of IUA in women of childbearing age with high risk factors.MethodsThe research methods was case-control study.From March 2015 to March 2016,80 patients with uterine adhesions were treated in our hospital's hysteroscopy center for the study group,used American Fertility Society classification as a diagnostic standard,Among them,22 patients with mild intrauterine adhesions,30 patients with moderate intrauterine adhesions and 28 patients with severe intrauterine adhesions;thirty cases of non-IUA patients with chronic salpingitis and intimal hyperplasia were selected as the control group.The age of both groups was 20-40 years old.Collected two groups of patients' endometrial tissue,the expressions of PDGF-AA and p-ERK1/2 in the endometrium were detected by immunohistochemical method,chi-square test and Kruskal-Wallis rank test were used to analyze the expression of PDGF-AA and p-ERK1/2 in the endometrium of the two groups.Results1.The expression of platelet-derived growth factor endometrial tissue : through immunohistochemical detection methods,the positive expression rate was 92.5%(74/80)in the study group,which was significantly higher than that in the control group 16.7%(5/30),the difference was statistically significant(P <0.001).The proportion of weakpositive(+)?positive(++)and strong positive(+++)in the study group was 36.3%(29/80)?31.3%(25/80)?25.0%(20/80),the proportion of weak positive(+)?positive(++)and strong positive(+++)in the study group was 13.3%(4/30)?3.3%(1/30)?0%(0/30),and the difference was statistically significant(P <0.001).The positive rates of PDGF-AA in the mild?moderate and severe intrauterine adhesions were 0%(0/22)?13.3%(4/30)? 57.1%(16/28),by chi-square test,the difference was statistically significant(P <0.001).That was with the degree of adhesion increased,PDGF-AA in the endometrial tissue expression increased.2.The expression of extracellular signal-regulated kinase 1/2 endometrial tissue :through immunohistochemical detection methods,the positive expression rate was95.0%(76/80)in the study group,which was significantly higher than that in the control group 26.7 %(8/30),the difference was statistically significant(P <0.001).The proportion of weak positive(+)?positive(++)and strong positive(+++)in the study group was 37.5%(30/80)?30.0%(24/80)?27.5%(22/80),the proportion of weak positive(+)?positive(++)and strong positive(+++)in the study group was 16.7%(5/30)?10.0%(3/30)?0%(0/30),and the difference was statistically significant(P <0.001).The positive rates of p-ERK1/2 in the mild ? moderate and severe intrauterine adhesions were 0%(0/22)?20%(6/30)?71.4%(20/28),by chi-square test,the difference was statistically significant(P <0.001).That was with the degree of adhesion increased,p-ERK1/2 in the endometrial tissue expression increased.3.Through the correlation analysis,the expression of p-ERK1/2 was closely related to the expression of PDGF-AA in the endometrium of patients with intrauterine adhesions(Kappa = 0.767,P = 0.000).Conclusions1.The expression of PDGF-AA and p-ERK1 / 2 may be related to the occurrence of intrauterine adhesions.2.With the degree of adhesion increased,PDGF-AA?p-ERK1/2 expression in theendometrium also increased,may be related to the development of intrauterine adhesions.3.PDGF-AA,p-ERK1/2 may be involved in the formation of intrauterine adhesions by interaction. |
