Objective:Chronic myeloid leukemia(CML)is a clonal disease of immune involvement.CD4+CD25+regulatory T cells(Treg cells)as a kind of negative regulator of immune cells shows different expression in chronic myeloid leukemia patients with different course.Analyse the expression of CD4+CD25+ regulatory T cells in CML patients can provide a clinical treatment plan to increase the efficacy of CML from the perspective of immunization.Methods:We collected the peripheral blood of 45 CML patients and 10 normal controls who were visited in the Northern Jiangsu People’s Hospital from July 2016 to February 2017.Flow cytometry was used to detect the proportion of CD4+CD25+regulatory T cells to T lymphocytes.The proportions was compared among the CML patients and normal controls、different courses of CML,to compare the expression level of CD4+CD25+Treg cells and its clinical significance.Result:1.There was a positive correlation between the proportion of CD4+CD25+regulatory T cells and the BCR-ABL fusion gene in CML(r=0.300998,P<0.05).2.Compared with normal controls,the frequency of CD4+CD25+ regulatory T cells in new diagnosed patients was significantly higher(P<0.05),and the frequency were significantly higher than those patients who didn’t reach MMR(P>0.05),while the frequency had no statistical significance in those patients who reach MMR(P<0.05);Compared with normal the new diagnosed patients,the frequency were decreased significantly after the control(P<0.05),especially in those who obtained MMR(P<0.05).3.The frequency of CD4+CD25+ regulatory T cells in blastic phase was significantly higher than those in chronic phase or accelerated phase(P<0.05),while there was no significant differencebetween chronic phase and accelerated phase(P>0.05).4.There was no significant difference of CD4+CD25+ regulatory T cells between the low-risk group and the intermediate-risk group(P<0.05),while the frequency was significant was higher in the high-risk group(P<0.05).Conclusion:There was a positive correlation between the frequency of CD4+CD25+ regulatory T cells and residual lesion、stage in patients、risk stratification with chronic myelogenous leukemia.The correlation applied that CD4+CD25+ regulatory T cells were involved in tumor immunosuppression.Removal of Treg cells from patients with CML may help to increase the efficacy of CML from an immune point of view.
