Objective:To observe the effect of puerarin on the hypoxia index and th e expression of Rho kinase in the model of hypoxic pulmonary hypertension rats,and to explore the possible mechanism of puerarin and provide the theor etical basis for the clinical application.Method:The rats were randomly divided into three groups:normal cont rol group,hypoxia model group,puerarin group and puerarin group.The rats were randomly divided into three groups:normal control group,hypoxia mod el group,puerarin The rats in the control group were cultured in the normal g roup under the normal environment,and the hypoxia group was cultured in th e hypoxic environment for 3 days,7 days,14 days and 21 days.The levels of pulmonary arterial pressure(mPAP),right ventricular hypertrophy index(R VHI),superoxide dismutase activity(SOD)and malondialdehyde(MDA)in 1 ung tissue were measured.The primary pulmonary artery smooth muscle cells(PASMC)were cultu red by collagenase digestion and the high CO2 hypoxia hypoxia model of rat pulmonary artery smooth muscle cells was established.(50?mol/L,100p,mo 1/L,200?mol/L)were randomly divided into model group and puerarin gro up.The rats were cultured in hypoxic environment and cultured for 24 h.The phosphorylation status of myosin phosphatase was detected by Western blot,which was used as a marker of activation of Rho kinase.The phosphorylatio n of myosin phosphatase was detected by immunofluorescence microscopy.Result:1.Compared with the normal control group,the mPAP of the hypoxia mo del group was significantly higher than that of the normal control group at 14 d and 21 d(P<0.05).Compared with the hypoxia model group,the mPAP o f the puerarin group was significantly increased at 14 d and 21 d(P<0.05).2.Right ventricular hypertrophy index results comparison:hypoxia model group rats right heart hypertrophy index of oxygen increases with time increased obviously,with significant difference(P<0.05);The index of dextral hypertrophy of the rats was also elevated,and the difference was significant(P<0.05).The increase of the 21 d numerical value of the ggenin group began to decrease evenly and was significantly different from the hypoxia group(P<0.01).3.Superoxide dismutase(SOD)numerical comparison:SOD activity in hypoxia model group rats significantly lower would be increased with the increase of oxygen time,starting from 14 d analysis found that data difference is significant(P<0.05);Puerarin treatment group rats has also a certain degree of superoxide dismutase(sod),14 d numerical comparison with hypoxia group,with significant difference(P<0.05);The reduction of the numerical value of the ggenin group 21 d began to decrease uniformly,and was significantly different from the hypoxia model group(P<0.01).4.Malondialdehyde content is the result of the comparison:oxygen malondialdehyde content in the model group rats would be increased with the increase of oxygen time increased significantly,14 d began measuring value compared with the normal control group,with significant difference(P<0.05);The levels of propylene glyaldehyde in the group of mice were also elevated to a certain extent,and the difference was significant(P<0.05).The numerical comparison of the data of the group 21 d in the treatment group of the ggenin treatment group began to slow,and the numerical comparison with the hypoxia model group was significantly different(P<0.01).5.The cultivation and morphology of PASMCs:the use of collagen digestion to train the success of the pulmonary smooth muscle cell is highly feasible.With inverted microscope microscopic observation:adherent cells grew well,cells were irregular triangle or long spindle,conforms to the physiological characteristics of pulmonary artery smooth muscle cells,are arranged in parallel or clusters of growth(table 1 a),grow 90%fusion,showed a typical "peak and valley" vascular smooth muscle cells grow characteristics(table 1 b).6.The effect of guggenin on hypoxia induced proliferation of PASMC:the optimal concentration of the ggenin intervention was 100 mu g/mL.Puerarin can significantly inhibit PASMC proliferation caused by hypoxia,so as to reduce hypoxia caused by pulmonary vascular remodeling,has guiding effect to the treatment of hypoxic pulmonary hypertension.7.The results of the Rho kinase test showed that the expression of Rho kinase in the model group was significantly higher after the lack of oxygen 24h,and the comparison with the model group was significantly different(P<0.05).The expression of the Rho in the concentration groups was significantly lower,with significant difference(P<0.01).Conclusions:Puerarin can effectively inhibit hypoxia caused by pulmonary artery mean pressure increasing and oxygen free radical,reduce the degree of pulmonary vascular remodeling,has good treatment effect to hypoxic pulmonary hypertension and prevention of protection.Puerarin have a protective effect on pulmonary arterial smooth muscle cells,for lack of oxygen causes of PASMC proliferation and pulmonary vascular reconstruction,Rho kinase levels caused by hypoxia,restrain and reduce the effect of puerarin is obvious,prove its on hypoxic pulmonary hypertension has the good protection and treatment effect.
