| Objective With the popularity of cervical cancer screening,the increased screening methods and techniques,the enhancement of women’s health consciousness,the detection rate of early cervical cancer is rising constantly,Surgery combined with neoadjuvant radiochemotherapy can achieve good results.However,the adverse reactions caused by this can seriously affect the quality of life of patients,and the therapeutic effect of neoadjuvant radiochemotherapy for advanced cervical cancer,drug resistance and recurrent cervical cancer is not very satisfactory,which has brought great difficulties to the clinical treatment.Therefore,it is of great significance to explore the mechanism of cervical cancer from the cellular and molecular level,and to find and conquer new targets for the treatment of cervical cancer,and we can isolate stem cells from various tissues by the side group(Side population,SP)phenotype.Therefore,the study of markers is a prerequisite for stem cell research,Previous research has shown that P63,ALDH1,ABCG2,P63 and CD44 may become cervical cancer stem cell markers,However,further studies are needed to confirm this.Aldehyde dehydrogenase 1(aldehyde dehydrogenase 1,ALDH1),a subtype of aldehyde dehydrogenase,mainly located in the cytoplasm,can catalytic oxidation of acetaldehyde to acetic acid.Which is expressed in normal cells,such as human hematopoietic stem cells and neural stem cells,as well as cancer stem cells,participating in the metabolism of many molecules in the cell and the differentiation of stem cells.ATP-binding cassettetransporter super family G2(ATP-binding cassette transporter super family G2,ABCG2),a multidrug resistance protein,belonged to ABC transport protein superfamily,is found to function as ATP-depended efflux pump transporting various drugs out of cell,and protect stem cells from being killed.However,whether ALDH1 and ABCG2 can be used as markers of cervical cancer stem cells remains to be confirmed,so far there have been no reports that whether the expression of ALDH1 and ABCG2 in cervical squamous cell carcinoma simultaneously and whether there is a correlation between the two expressions.SP immunohistochemistry method was used in this study to detect the expression of ALDH1 and ABCG2 in 76 cases of cervica squamous cancers,52 cases Of HSIL,and 37 cases of chronic cervicitis tissues,and analyze the results,and investigate whether the expression of two markets in cervical cancer is correlated and the relationship between the expression of two and clinicopathological factors in cervical carcinoma,to explore the possibility whether ALDH1 and ABCG2 can be regarded as markers of cervical squamous cell stem cells,and to provide clues and experimental basis for the study of cervical cancer stem cells and targeted therapy of cervical cancer.Methods Patients pathologically confirmed 76 cases of cervical squamous cell carcinoma,receiving radical hysterectomy of cervical cancer,52 cases of high grade squamous intraepithelial lesions(HSIL),receiving cervical biopsy or cervical conization or hysterectomy,who were admitted to the First Affiliated Hospital of Zhengzhou University from December 2012 to December 2014 were collected,selected the corresponding period 37 cases patients underwent total hysterectomy because of uterine fibroids in the hospital and cervical pathology pathological diagnosis with chronic cervicitis as control.The age of patients with cervical squamous cell carcinoma ranged from 37 to 64 years old,with an average age of 52 years;according to the International Federation of gynecology and Obstetrics(FIGO,2009)clinical staging standards,including 51 cases of stage I,25 cases of stageⅡ.According to the degree of differentiation of cervical cancer WHO,G1,G2(high,moderate differentiation)42 cases,G3(low differentiation)level 34 cases.All patients had complete medical records and were not treated with radiotherapy,chemotherapy or biological therapy before taking the specimen.SP immunohistochemistry method was used to detect the expression of ALDH1 and ABCG2 in the paraffin tissue sections.Phosphate buffer(PBS)in place of a resistance was used as a negative control set up at each staining.Results 1.The positive expression rates of ALDH1 and ABCG2 in chronic cervicitis tissues were 5.41%(2/37)and 8.11%(3/37),HSIL tissues were 23.08%(12/52)and 28.85%(15/52),cervical cancers tissues were 53.95%(41/76)and 61.84%(47/76),Their positive expression rates in cervical cancers tissues were significantly higher than in HSIL,and Chronic cervicitis tissues,with statistical significance(P<0.05).2.The positive expressions of ALDH1 in cervical cancer tissues were showed correlation with tumor grade and lymph-node metastasis(P<0.05),The positive expressions of ABCG2 in cervical cancer tissues were showed correlation with tumor grade,clinical stage and lymph-node metastasis(P<0.05).3.The expressions of ALDH1 and ABCG2 in cervical cancer were positive relative(r=0.535,P<0.05).Conclusion 1.The increased expressions of ALDH1 and ABCG2 in cervical squamous cancer,have relationship to the differentiation of tumor grade,the two markets may play a role in the occurrence,development of cervical squamous cell carcinoma.2.The increased expressions of ALDH1 and ABCG2 in cervical squamous cancer were showed correlation with lymph-node metastasis,the two markets may be closed related to the metastasis of cervical squamous cell carcinoma.3.The expressions of ALDH1 and ABCG2 in cervical cancer were positive relative,they might have some kind of synergistic effect in occurrence,development,lymphatic metastasis of cervical squamous cell carcinoma. |
