objective: To explore The effect and related mechanism of 1,25-(OH)2-VitD3 on secretion of extracellular matrix.Method:Screen out the most efficient SiRNA,and the optimum composition ratio of the plasmid and the liposome.Cells were divided into 6 groups: cells were treated with SiRNA,cells were treated with blank vector,cells were treated with PBS in normal control group,cells were treated with SiRNA and VitD3,cells were treated with blank vector and VitD3,normal control group cells treated with VitD3.The gene levels of VDR、TGF-β1、Smad3、Smad7、FN were evaluated using reverse transcription polymerase chain reaction(RT-PCR).Result: The gene levels of TGF-β1、FN were inhibited obviously after treated with VitD3(P<0.05).And this effect went down if the VDR gene were knocked out.Treated with VitD3 could also down-regulate the expression of Smad3 and up-regulate the expression of Smad7,this effect could be declined with the knocked out of the VDR gene.Conclusion: VitD3 can inhibit the mRNA expression of TGF-β1 and FN.The mechanism may rely on the combine of VitD3 and VDR,and its effect on regulate the expression of TGF-β1 may be related to the inhibition of Receptor-regulated Smads and the excitation of Antagonistic Smads.
