| Part One The Establishment of Animal Model of Non-alcoholicSteatohepatitis In Wistar RatsObjective:To establish an animal model of non-alcoholic steatohepatitis in(Wistar)rats,and to provide an experimental animal model to study the protective effect and mechanism of saturated hydrogen saline on non-alcoholic steatohepatitis.Methods:Using high-fat and high-cholesterol feed Wistar rats for 4-18 weeks.From the beginning fourth week,weekly observe the appearance of steatosis and inflammatory cell infiltration and the level of fatty degeneration and inflammatory cell infiltration,μltimately determine the appropriate feeding time.NASH rat model was successfμlly constructed for subsequent research.Results:At fourth weeks,There were no fat deposition in the liver in high fat diet group microscopically,but mild local fat deposition were exist,at eighth weeks showed microscopically diffuse fat deposition and local small amount of inflammatory cell infiltration,at sixteenth weeks,there was microscopically infiltration of inflammatory cells in the liver,and punctiform necrosis.ALT in high fat diet group and normal diet group showed significant differences at eighth and sixteenth weeks,while AST in high fat diet group and normal diet group showed significant differences in fourth,eighth and sixteenth weeks.the serum total cholesterol(TC)and triglyceride(TG)in high fat diet group increased gradually,and there were differences compared with normal diet group respectively at fourth,eighth and sixteenth weeks.Conclusion:Feed Wistar rats with high fat and high cholesterol diet to replicate animal model in non-alcoholic steatohepatitis,stable non-alcoholic steatohepatitis occurred at sixteenth weeks.Part Two The Protective Effect of Hydrogen Saline on Non-alcoholic SteatohepatitisObjective : To prove that the treatment effect of hydrogen on non-alcoholic steatohepatitis in rats and to identify the concrete molecμlar mechanism.Methods:Wistar rats were randomly divided into two groups: treatment group and control group,the treatment group was treated with saturated hydrogen saline,control group was treated with normal saline.Before and after treatment in 24 hours,1 week and 4 weeks,peripheral blood biochemical indexes(transaminase,blood lipid and blood glucose),liver morphological changes,oxidative stress,inflammatory factors,apoptosis and signal transduction pathway were detected in two groups.Resμlts : The cholesterol and triglycerides in treatment group and experimental group were continued to increase after giving hydrogen saline,but there were no significant difference during the same observation time,ALT and AST did not differ between the control group and the experimental group at first days after treatment,but there were significant difference between the two groups at first and fourth weeks.The expression of IL-6 and TNF also did not differ at first days before and after treatment,but did obviously differ at first and fourth weeks.There were no significant difference in the expression of caspase-3 in the two group at first days before and after treatment and week first,but in the fourth weeks,the expression in control group was significantly higher than that in the experimental group.Conclusion:As a kind of selective antioxidant,Saturated hydrogen saline has a certain protective effect on NASH. |
