Study On The Expreession Of AQP3 And AQP8 After Subtotal Colectomy In Rats And The Effect Of Drugs

Objective: Through the observation of AQP3,8 expression in residual rat colon after hemicolectomy and the influence of drugs to them,in order to researches the remaining colon whether there is a compensatory increase in expression of aquaporin 3,8 to enhance colon water absorption.And Validation of Tongxieyaofang,loperamide hydrochloride can promote the water channel protein(Aquaporin AQP)the role of 3,8 expression or not.Therefore,we can provide the method for the clinical diagnosis and treatment of diarrhea after hemicolectomyMethods: Select 36 adult male SD rats,randomly divided into the control group(group A),model group(group B),Tongxieyaofang group(group C),loperamide hydrochloride group(group D),each group of 9 rats.Among them,the blank group A,single cage conventional breeding;B-D groups were performed under general anesthesia partial resection and intestinal anastomosis oprations,then single cage of conventional breeding and anti-infection treatment 3 days;Group C,group D were intragastric administration one time everyday maintaining one week with Tongxieyaofang,loperamide hydrochloride.Respectively,after 1,2,and 4 weeks,drawn the colon specimen from the same part of each rat which was selected randomly from those groups.Then detected the expression of colon AQP3、8 based on immunohistochemistry and real-time quantitative polymerase chain type reaction(Real-Time RT-PCR)methods and requirements,finally made a comparison of corresponding results by the appropriate statistical methods.Results: 1.Compared with A group,the expression of AQP3,8 in the remaining colon tissue of B group,C group and D group were increased,the difference was statistically significant(p <0.01).2.Compared with B group,the expression of AQP3,8 in the remaining colon tissue of C group and D group were increased,the difference was statistically significant(p <0.05).3.Compared with D group,the expression of AQP3,8 in the remaining colon tissue of C group were increased,the difference was statistically significant(p <0.05).Conclusion: The expression of AQP3,8 in the residual colon after subtotal colectomy was up-regulated,and the activity was correlated with the postoperative time;Tongxieyaofang and loperamide hydrochloride had a significant role in promoting the expression of AQP3,8 in the remaining colon,and the promoting effect of the Tongxieyaofang is more obvious.
