| Objective:To investigate the effects and mechanisms of sodium tanshinoneⅡA sulfonate(STS)on pirarubicin(THP)induced acute cardiotoxicity.Methods: Sprague-Dawley female rats were randomly divided into four groups and 10 rats in each group as follows: group control(GNS),group THP,group STS-1 and group STS-2.Glucose Saline(GNS)dose of 10ml/kg in rats of group control were intraperitoneal injection once a day for seven days.GNS dose of 10ml/kg were intraperitoneal injection for seven days in rats of group THP,and GNS were injection in 30 min later on the fifth days,THP dose of 15mg/kg were intraperitoneal injection by once finished in 2min.STS dose of 10mg/kg were given once a day for seven days in rats of group STS low dose(STS-1);and low dose of STS were injection in 30 min later on the fifth days,THP dose of 15 mg/kg were intraperitoneal injection by once exposure finished in 2min.STS dose of 20 mg/k were given in rats of group STS high dose(STS-2)once a day for seven days;high dose STS were injection in 30 min later on the fifth days,THP dose of 15 mg/kg were intraperitoneal injection by once exposure finished in 2min.All rats were anesthetized by chloral hydrate on the eighth day of the experiments,and blood plasma were collected by abdominalaortic,enzyme-linked immunoabsorbentassay(ELISA)methods were applied in the detection of serum troponin-I(cTnI)level,N-Terminal Pro-Brain Natriuretic Peptide(NT-proBNP)level,hyper-sensitive C-reactive protein(hs-CRP)and homocysteine(Hcy).The hearts of rats were harvested and weighted.Pathologic structures were detected and scored in a blinded way.Carotid arteries and left ventricular free walls(FW)were stained respectively by hematoxylin and eosin(HE).Similarly,to observe the characteristics of pathologic structures and grade;The rest cardiac tissues were used to made homogenate to observe the change and activities of MDA and SOD.Results:(1)Compared with GNS、STS-1 and STS-2 group,the rats in THP group showed some symptoms such as listlessness,little activity,loss of appetite,depilation and diarrhea.(2)Weights :There were no difference in weights of the rats in four groups before any treating.When THP were administered for three days post exposure,weights in rats of group THP were less than that significantly compared with group GNS,group STS-1and group STS-2(P<0.01);and that had no statistical significance in another rats of three groups were compared with each other(P>0.05).(3)Heart weight index(HWI)in rats of THP group were prominently higher than that in other three groups with statistical significance(P<0.05).HWI were significant difference in rats between group STS-1 and group STS-2(P<0.05).(4)The serum level of cTnI and NT-pro BNP: These of rats in group THP had obvious difference with other three groups(P<0.05).There were no statistical significance were compared with group GNS,group STS-1 and group STS-2 in pairs(P<0.05).(5)The serum hs-CRP and Hcy:These of rats in group THP had showed significant difference with that in other three groups(P<0.05).The serum level of hs-CRP and Hcy were improved significantly were compared with group GNS,groupSTS-1 and group STS-2(P<0.05).There were significant difference between group STS-1 and group STS-2(P<0.05).(6)SOD and MDA: the activities of SOD in group THP were significantly lower than that in group GNS,group STS-1 and group STS-2(P<0.05).But these were showed no statistical significance between group GNS,group STS-1 and group STS-2(P>0.05).The level of MDA in group THP were obviously higher than that in other three groups(P<0.05).But these no significant difference between group GNS,group STS-1 and group STS-2(P>0.05).(7)Microscopic observation: The observations of carotid arteries and left ventricular showed that the injury of group THP were severer than that in group STS-1 and group STS-2.Cardiac pathology Scores: these in group THP were significantly higher than that in group GNS,group STS-1 and group STS-2(P<0.05).And these were difference significantly between group GNS,group STS-1 and group STS-2(P<0.05).Conclusion: STS can improve the activities of antioxidant enzymes,decrease the lipid peroxidation and inflammatory reaction,which can alleviate myocardial injury and protective effect. |
