| Objective: To detect the expression of β-catenin,Wnt3 a and Lgr5 in different cervical tissues,and analyze the correlation among the expression,to explore their relationship with the occurrence and development of cervical cancer.Methods: The expression of β-catenin,Wnt3 a and Lgr5 in different cervical tissues were detected by immunohistochemical method(SP method),including 61 cases of cervical cancer tissue,30 cases of cervical intraepithelial neoplasia(12 cases of CINI,18 cases of CINII-III),19 cases of normal cervical tissue.Results:1.Under normal conditions,the expression of β-catenin was continuous in the cell membrane,the pathological state,there is a loss of membrane expression,and a different level of abnormal expression in cytoplasmic/nuclear.The abnormal expression rate of β-catenin was 5.26%(1/19)in normal cervical tissue,33.33%(4/12)in CINI group,38.89%(7/18)in CINII-III group and 75.41% 61),comparison between 4 groups(χ2=32.958,P <0.05),with statistically significant.The abnormal expression rates of G1,G2 and G3 grade were 41.67%(5/12),76.67%(23/30),94.74%(18/19)in cervical cancer,It can be seen that it is prominent in G3 group expression,(χ2=11.221,P <0.05),with statistically significant.The abnormal expression rates in the shallow infiltration group and the deep infiltration group were 55.56%(10/18)and 83.72%(36/43)(χ2=5.428,P <0.05),with statistical significance.The abnormal expression may not relate to clinical stage,age,histological type and age(P >0.05).2.The positive expression rate of Wnt3 a was 10.53%(2/19)in normal cervical tissue group,41.67%(5/12)in CINI group,55.56%(10/18)in CINII-III group,40.98%(25/61),comparison between 4 groups(χ2=8.723,P <0.05),with statistically significant.Its’ positive expression rate was 59.09%(13/22)in lymph node metastasis group,30.78%(12/39)in no lymph node metastasis group,with statistically significant(χ2=4.665,P< 0.05),and Wnt3 a positive expression may have no statistically significant in clinical stage,Infiltration depth,histological type,age(P>0.05)3.The positive expression rate of Lgr5 was 0%(0/19)in normal cervical tissue group,25%(3/12)in CINI group,44.44%(8/18)in CINII-III group,65.57% in group CC(40/61),comparison between 4 groups(χ2=28.130,P<0.05),with statistically significant.The positive expression of Lgr5 in G1 33.33%(4/12),G2 70%(21/30)and G3 70%(5/19)levels was statistically significant(χ2=7.291).The abnormal expression rates were 38.89%(7/18)and 76.74%(33/43),in the shallow infiltration group and the deep infiltration group,with statistically significant.and the positive expression of Lgr5 in clinical stage,pathological grade,infiltration depth,histological type and age was with no statistically significant(P>0.05).4.In CC group,for the expression of β-catenin and Wnt3 a,there were 23 cases of both positive expression,13 cases of both negative expression,have a positive relationship(r=0.321,P <0.05).for the expression of β-catenin and Lgr5,there were 35 cases of both positive expression,10 cases of both negative expression,have a positive relationship(r=0.321,P<0.05).for the expression of Wnt3 a and Lgr5,there were 13 cases of both positive expression,9 cases of both negative expression,with no statistically significant(P>0.05).Conclusions:1.The abnormal expression of β-catenin,wnt 3a and Lgr5 in normal cervical tissues,CINI CINII-III and CC showed a gradual increase,suggesting that β-catenin,Wnt 3a and Lgr5 may affect the development of cervical cancer.2.Wnt3 a may be related to the occurrence of Precancerous lesions and the transfer of cervical cancer,β-catenin and Lgr5 may be related to the degree of malignancy and invasion of cervical cancer.3.There was a positive correlation between the expression of Lgr5 and β-catenin,wnt 3a and β-catenin,suggesting that the expression of Lgr5 and wnt 3a may activate the wnt signaling pathway,leading to β-catenin move to cytoplasmic / nuclear,and cause abnormal cell proliferation and the occurrence of cervical cancer. |
