Objective: Detection of HER-2 gene amplification by TMA and FISH in breast cancer,including 30 patients with unilateral breast cancer and 30 cases of bilateral primary breast cancer,discuss the prognosis.Methods: Collect 30 cases of unilateral breast cancer and 30 cases of bilateral primary breast cancer in jiangxi province tumor hospital from January 2003 to January 2010(paraffin-embedded tissues of invasive ductal carcinoma),detection of HER-2 gene amplification by TMA and FISH in all cases.Results:1.In 30 patients with unilateral breast cancer,HER-2 was detected in 9 cases,amplification rate of 30%.In 30 cases of bilateral primary breast cancer patients,HER-2 was detected in 2 cases,amplification rate of 3.3%.The two results showed significant correlation(P < 0.01).2.In 30 patients with unilateral breast cancer,3-years overall survival rate was 73%;in 30 cases of bilateral primary breast cancer,3-years overall survival rate was 93%;The two results showed significant correlation(P < 0.05).In 30 patients with unilateral breast cancer,5-years overall survival rate was 60%;in 30 cases of bilateral primary breast cancer,5-years overall survival rate was 73%;The two results showed significant correlation(P < 0.05).Conclusion: The combining application of TMA and FISH technology could detect the amplification of HER-2 gene successfully and exactly.HER-2 gene in patients of bilateral primary breast cancer expression rate is lower than the patients of unilateral breast cancer.The prognosis of patients with bilateral primary breast cancer is not worse than unilateral breast cancer patients,HER-2 as independent factor,excessive expression in breast cancer patients with poor prognosis.
