Objective: Luteoloside(Lut),a kind of flavonoids found in plenty number of Chinese herbs,exerts myocardial protection effects.To investigate the protective effects of Lut on cardiomyocytes which were exposed to A/R injury and explore whether the cardioprotection of Lut is related with the expression of 14-3-3η,in our study,H9c2 cells are pretreated with Lut,subsequently undergone anoxia/reoxygenation(A/R)treatment.Methods: In our study,H9c2 cardiomyocytes pretreated with Lut were suffered from A/R injury model.MTS assay was applied to analyze the cell viability;the level of lactate dehydrogenase(LDH)was tested by spectrophotometer;Western Blot assay was conducted to examine the protein expressions of 14-3-3η;flow cytometry was used to detect the intracellular reactive oxygen species(ROS);spectrophotometer was used to detect the openness of mitochondrial permeability transition pores(mPTP)and activity of caspase-3;Western Blot assay was conducted to examine the protein expressions of Bcl-2,and Bax;flow cytometry was used to detect the cell apoptosis.Results: H9c2 cells of each group were treated with A/R injury.1.Lut pretreatment increased the cell viability and decreased the activity of LDH;2.Lut pretreatment increased the expression of 14-3-3η protein;3.Lut pretreatment reduced the ROS production;4.Lut pretreatment inhibited the opening of mPTP;5.Lut pretreatment restrained the activity of caspase-3;6.Lut pretreatment enhanced the expression of Bcl-2 and decreased the expression of Bax protein;7.Lut protected H9c2 cells against A/R-induced apoptosis.The results above were all statistically significant(p < 0.01).Conclusion: Luteoloside protects H9c2 cardiomyocytes against A/R injury by upregulating 14-3-3η protein,and the cardioprotective effect involves mitochondrial pathway.
