Trichosanthes kirilowii Maxim is a flowering plant in the Cucurbitaceae family,which is widely used in traditional Chinese medicine.This paper studied the effects of ultrasonic extraction and matrix solid phase extraction on the extraction of fatty acids in Trichosanthes kirilowii by high sensitivity fluorescent probe and reverse reverse phase high performance liquid chromatography(HPLC).The results showed that the matrix solid phase extraction method can be usaed as a stable and rapid analysis of fatty acids content of the content of fatty acids in Trichosanthes kirilowii Maxim.The reagent,2-(9-oxoacridin-10(9H)-yl)acetohydrazide(OAAH),has been synthesized and successfully applied to the HPLC determination of fatty acids of Trichosanthes kirilowii Maxim.The derivatives exhibit excellent fluorescence property with excitation and emission wavelengths of 260 nm and 430 nm,respectively.The 20 derivatives of fatty acids were separated on a BDS C8 reversed-phase column with gradient elution.Good linear correlations were observed for all fatty acids with correlation coefficients of >0.996.Free fatty acids in the roots,stems,leaves and peels of Trichosanthes kirilowii Maxim were analyzed by the developed method.An efficient method for the extraction and determination of free fatty acids(FFAs)in the whole Trichosanthes kirilowii Maxim plant was developed based on matrix solid-phase dispersion(MSPD)extraction.MSPD extraction was performed using aminopropyl-functionalized SBA-15(N-SBA-15)to extract FFAs via acid–base interactions.The optimum parameters for the MSPD extraction were N-SBA-15 30 mg,grinding time 120 s,and elution solvent methanol–HCl(6:4,v/v).The content of each component measured by matrix solid-phase dispersion method and the ultrasonic extraction method were within 5%.The results showed that this solid phase extraction method for fatty acid extraction improved the extraction efficiency of fatty acid and simplified the extraction process.The phenolic acids in Trichosanthes kirilowii were extracted by ultrasonic extraction method,and the separation of PA components was performed on a Hypersil Gold C18 reverse phase column with gradient elution.The separation 8 kinds of phenolic acids were completed and this method was used for the determination of phenolic acids in Trichosanthes kirilowii Maxim.
