Serum albumin is the carrier for storage and transport of small molecule drugs.Study the mechanism of the interaction between drug and protein has important significance for clarifying the pharmacology and pharmacokinetic behavior.Therefore,the interaction studies between drug and protein have become an important research topic span chemistry,biology and clinical pharmacy.In this article,the interaction between four flavones and bovine serum albumin and the influence of metal ions were investigated under physiological condition in vitro.On the basis of the previous research,the study were carried out using fluorescence spectroscopy,synchronous fluorescence spectroscopy,three-dimensional fluorescence spectroscopy,U V absorption spectroscopy,circular dichroism.In order to provide the physiological effects information of rare earth metals,we have studied the interaction with bovine serum albumin and three rare earth metals.The main contents are as follows:1.The interaction of four structurally related flavonoids(hesperetin,Diosmetin,luteolin,quercetin)and bovine serum albumin were studied by multi-spectral method.Conclusion was rearched by comparing the quenching constant,the binding constant,the number of binding sites,the distance and the force type,protein conformational changes,etc.Structure-activity relationship of flavonoids derived through analyzing the effects of C2-C3 single and double bonds,adjacent hydroxyl hydrogen of B ring,3-hydroxyl of C ring.These resulates could be used to guide the development and transformation of flavonoids drugs.2.Using fluorescence spectroscopy and UV absorption spectroscopy to study the characteristics of flavonoid-Me-protein system in the presence of metalions(Cu2+、Zn2+、Ca2+、Al3+)in vitro conditions.The effects of metal ions on inter molecular interactions were reached by comparing the combine parameters and conformational changes of flavonoid-protein system and flavonoid-Me-protein system.It could be used for guiding clinical safety and rational use of drugs.3.Using fluorescence spectroscopy and UV absorption spectroscopy to study the interactions between bovine serum albumin and lanthanide rare earth ions(Ce3+,Nd 3+ and Yb3+).It revealed the interaction law of rare earth ions on the organism and provided molecular-level information for pharmacodynamic and toxicological studies.
