Objective: The purpose of this study is to evaluate the potential neuroprotective effect of leonurine and underlying mechanisms after cerebral ischemia.Methods: 1.Clinical experiment: Patients with cerebral ischemic shock were randomly divided into conventional treatment group(n=60)and conventional treatment plus leonurine treatment group(n=60).The serum levels of oxidative stress markers and therapeutic effect were compared.2.Animal experiment: The SD rats were randomly divided into control group,cerebral ischemic shock group,leonurine-treated group and leonurine-treated+ PI3K/Akt blocked group.The levels of oxidative stress markers and pathological changes in brain tissues were observed.Results: Compared with control group,the serum levels of SOD、GPx、CAT、TrxR were decreased in cerebral ischemic shock group,and MDA was elevated(P<0.05).Treatment with leonurine effectively elevated the levels of the above antioxidant markers(P<0.05).And inhibiting the expression of PI3K/Akt pathway could partly reduce the beneficial effect of leonurine cerebral ischemic shock(P<0.05).In patients,treatment with leonurine had the same effect,which were demonstrated by elevated levels of anti-oxidative stress markers and faster recovery of patients(P<0.05).Conclusions: Oxidative stress injury played an important role in genesis of cerebral ischemic shock.Treatment with leonurine could effectively ameliorate oxidative stress response,facilitate the neural functional recovery.Its effect was partly mediated by PI3K/Akt pathway.Treatment with Bushentongmai recipe,which main component is leonurine,could improve the clinical efficacy when it was used to treat cerebral ischemic shock.
