Object: To explore the association of GSTP1(rsl695) gene polymorphism with susceptibility and clinicopathologic characteristics of breast cancer in GuangXi population.Methods: Polymerase chain reaction-restriction fragment length polymorphism(PCR-RFLP) and Case-control study were adopted to detect the gene polymorphism and investigate the distribution of GSTP1(rs1695) in 230 breast cancer patients and in 230 healthy persons in GuangXi province. We analysis the association of GSTP1(rs1695) gene polymorphism with susceptibility and clinicopathologic characteristics of breast cancer by statistical methods.Results:1.Hardy-Weinberg equilibrium genetic testingGenotype distributions of GSTP1(rs1695) loci in both groups population were consistent with Hardy-Weinberg equilibrium2.1n general population, risk evaluation of the GSTP1(rs1695) genotypes and alleles on the development of breast cancer.In general population, the genotype frequencies distributions of A/A, A/G and G/G were -significantly different between cases and controls groups (P<0.05) the genotypes frequency of variant genotypes (A/G+G/G) was statistically higher in the breast cancer group compared with the controls with significantly different(P<0.05).In A/A genotype as a reference, The risk to breast cancer was respectively increase 0.941 times (OR=1.941, 95%CI=1.290-2.921, P=0.001) and 0.799 times(OR=1.799, 95%CI=1.216-2.662,P=0.003) among A/G genotype individual and females carrying variant genotypes (A/G+G/G). A and G alleles distributions were significantly different between two groups (P<0.05), and the oddsrations of G alleles was 1.497(95%CI=1.071-2.093, P=0.018) with respect to A alleles.3.In ethnic Zhuang population, Risk evaluation of the GSTP1(rs1695)genotypes and alleles on the development of breast cancerIn ethnic Zhuang population,the genotype frequencies distributions of A/A,A/G and G/G were significantly different between cases and controls groups(P<0.05) the genotypes frequency of variant genotypes (A/G+G/G) was statistically higher in the breast cancer group compared with the controls with significantly different(P<0.05). In A/A genotype as a reference, The risk to breast cancer was respectively increase 2.100 times (OR=3.100, 95%CI=1.369-7.020, P=0.007) and 1.750 times (OR=2.750, 95%CI=1.270-5.954,P=0.010) among A/G genotype individual and females carrying variant genotypes (A/G+G/G). In ethnic Zhuang population, A and G alleles distributions were significantly different between two groups (P<0.05), and the oddsrations of G alleles was 2.087(95%CI=1.066-4.083,P=0.032) with respect to A alleles.4.1n Han population, Risk evaluation of the GSTP1(rs1695) genotypes and alleles on the development of breast cancer.In Han population, the genotype frequencies distributions of A/A, A/G and G/G were significantly different between cases and controls groups (P<0.05) the genotypes frequency of variant genotypes (A/G+G/G) was statistically higher in the breast cancer group compared with the controls with significantly different(P<0.05).In A/A genotype as a reference, The risk to breast cancer was respectively increase 0.967 times (OR=1.967, 95%CI=1.168-3.312, P=0.011) and 0.851 times (OR=1.851,95%CI=1.125-3.045,P=0.015) among A/G genotype individual and females carrying variant genotypes (A/G+G/G). In Han population, A and G alleles distributions were significantly different between two groups (P<0.05), and the oddsrations of G alleles was 1.559(95%CI=1.019-2.383, P==0.041) with respect to A alleles.5.The association of the distribution of the GSTP1 (rs1695) genotypes with the clinicopathologic characteristics of breast cancerWhen stratifying by the clinical stage and histological type of breast cance, we found no statistical association (P>0.05).Conclusion:1.GSTPl(rs1695) polymorphisms are associsted with breast cancer susceptibility in GuangXi population, and variant genotypes (A/G+G/G) may notably increased the risk for breast cancer.2.GSTP1(rs1695) G alleles was a risk factor for breast cancer with respect to A alleles, and it may notably increased the risk of breast cancer.3.By stratifying analysis for ethnics, we found that variant genotypes(A/G+G/G) may notably increased the risk of breast cancer both in ethnic Zhuang and in Han population, and the risk was higher obviously Zhuang population than Han population, both of them higher than general population.4.GSTP1(rs1695) polymorphisms was not significantly associsted with the clinical stage and histological type of breast cance,5.Genotype distributions of GSTP1(rs1695) loci in both groups population were consistent with Hardy-Weinberg equilibrium, indicating that it has a good representativeness of the population.
